ETR1-allelic constitutions of autochthonous apple genotypes

dc.description.abstractEtilen je najjednostavniji biljni regulator rastenja koji ima krucijalnu ulogu u dozrevanju klimakteriĉnog ploda jabuke (Malus × domestica Borkh.). U percepciju etilena ukljuĉena je familija receptora, locirana na membrani endoplazmatiĉnog retikuluma. ETR1 pripada potfamiliji I receptora za etilen kod jabuke. Cilj ovog rada je da se ispita alelni polimorfizam ETR1 gena kod 48 autohtonih genotipova jabuke, koji se nalaze u ex situ kolekcionom zasadu na objektu Zdravljak Instituta za voćarstvo, Ĉaĉak. Alelni polimorfizam ETR1 gena je utvrĊen posle digestije, lanĉanom reakcijom polimeraze (PCR metodom) umnoţenog genomskog fragmenta sa tri restrikciona enzima - RsaI, AluI i HinfI. PoreĊenjem polimorfizma uoĉenog posle restrikcije (RsaI - fragmenti veliĉine 800 i 890 bp; Alu I - polimorfni fragment veliĉine 850 bp; HinfI - fragmenti veliĉine 800 i 1.130 bp), identifikovana su ĉetiri alela ETR1 gena - a, b, c i d u prouĉavanom autohtonom materijalu. Na osnovu utvrĊenih alelnih konstitucija ETR1 gena, ispitivani genotipovi se mogu svrstati u ĉetiri grupe (aa ‒ 27 genotipova; b,a/c ‒ 9 genotipova; c,a/c ‒ 3 genotipa; d,a/c/d ‒ 9 genotipova). Alel d ETR1 gena je do sada identifikovan iskljuĉivo kod autohtonih genotipova: ‘Bаbоvаčа’, ‘Bоšnjаnkа’, ‘Funtаčа (Оpаlјеnik)’, ‘Kаmеnicа 1’, ‘Оvčiјi nоs’, ‘Strеkinjа’, ‘Šаrеnikа’, ‘Тip 3’ i ‘Тip 6’. Molekularnom karakterizacijom i prоuĉаvаnjem nајvаţniјih biоlоških оsоbinа autohtonih genotipova, dаćе se znаĉајаn dоprinоs оĉuvаnju gеnеtiĉkе divеrgеntnоsti gеnоfоndа jabuke, оdnоsnо оbоgаtićе se kоlеkciјa pоĉеtnоg mаtеriјаlа kоја zаuzimа klјuĉnо mеstо u stvаrаnju nоvih sоrti.sr
dc.description.abstractEthylene is the simplest plant growth regulator which plays a crucial role in the ripening of climacteric apple (Malus × domestica Borkh.) fruit. The family of endoplasmic reticulum-localized receptors is involved in ethylene perception. ETR1 belongs to subfamily I of ethylene receptors in apple. This work was undertaken primarily to study the allelic polymorphism of the ETR1 gene in 48 autochthonous genotypes from the ex situ apple collection at Zdravljak facility of the Fruit Research Institute, Čačak. Polymorphisms were detected by digestion of PCR product with three restriction enzymes - RsaI, AluI and HinfI. The comparison of polymorphisms observed upon the restriction analysis (RsaI – two segregating fragments of 800 and 890 bp; AluI – one polymorphic fragment of 850 bp; HinfI – two segregating fragments of 800 and 1,130 bp) resulted in the identification of four alleles of ETR1 gene - a, b, c and d in the assessed autochthonous material. Based on obtained ETR1 allelic constitutions, the autochthonous genotypes were grouped into four classes (aa ‒ 27 genotypes; b,a/c ‒ 9 genotypes; c,a/c ‒ 3 genotypes; d,a/c/d ‒ 9 genotypes). So far, allele d has been identified only in the following autochthonous genotypes: ‘Bаbоvаčа’, ‘Bоšnjаnkа’, ‘Funtаčа (Оpаlјеnik)’, ‘Kаmеnicа 1’, ‘Оvčiјi Nоs’, ‘Strеkinjа’, ‘Šаrеnikа’, ‘Тip 3’ and ‘Тip 6’. Molecular characterization and study of the most important biological traits of autochthonous genotypes can significantly contribute to preservation of the genetic divergence in the apple germplasm. In addition, the genotypes with useful traits can provide the increase in the number of genotypes with a potential to be used as parents in developing new apple cultivars.sr
