The influence of imidazole fungicide on multiplication in vitro of low vigorous sweet cherry rootstock Gisela 6

U radu je ispitivan uticaj imidazol fungicida na multiplikaciju slabo bujne podloge za trešnju Gisela 6, samostalno, ili u kombinaciji sa N6-benzil-aminopurinom (BAP). Kao izvor imidazol fungicida korišćen je komercijalni preparat SPORTAK 45 - EC - 450 čija je aktivna supstanca N-propil-N-[2-(2,4,6-trihlorfenoksi)-etil]- imidazol-1-karboksiamid (prohloraz). U eksperimentu je ispitano 19 tipova medijuma u fazi multiplikacije, sa makro i mikro solima po Murashige i Skoog (1962) (MS), ali sa različitim koncentracijama prohloraza (PRO), BAP, IBA i GA3. Prohloraz je korišćen u 3 koncentracije (1, 5 i 10 μM), samostalno, ili u kombinaciji sa BAP (4,4 μM), IBA (5 μM) i GA3 (0,3 μM). Posle druge supkulture utvrđivani su parametri multiplikacije izdanaka, indeks multiplikacije, dužina i broj listova osovinskog i bočnih izdanaka. Takođe je merena sveža i suva masa izdanaka. Najveći indeks multiplikacije (1:4,94), kao i sveža/suva masa izdanaka dobijeni su na medijumu koji je sadržavao 5 μM PRO, ...4,4 μM BAP i 0,3 μM GA3. Primena fungicida samostalno, kao i u kombinaciji sa IBA ili GA3 nije imala značajan uticaj na multiplikaciju podloge za trešnju Gisela 6, bez obzira na upotrebljenu koncentraciju PRO. Međutim, sve tri ispitane koncentracije prohloraza u kombinaciji sa IBA uticale su na rizogenezu, pri čemu je najveći procenat ožiljavanja dobijen na medijumu sa 10 μM PRO (83,33%). Dobijeni rezultati pokazuju da PRO pojačava efekat egzogeno dodatog BAP na multiplikaciju vegetativne podloge za trešnju Gisela 6, a u kombinaciji sa IBA utiče i na rizogenezu.
The effect of imidazol fungicides applied either individually or combined with N6-benzyl-aminopurine (BAP) on the multiplication of low vigorous sweet cherry rootstock Gisela 6 was studied in the paper. The commercial chemical SPORTAK 45 - EC - 450 containing N-propyl-N-[2-(2,4,6-trichlorphenoxy)-ethyl] - imidazole-1-carboxamide(prochloraz) as an active substance, was used as the source of imidazole fungicide. Some 19 medium types were studied over the multiplication phase. The media, containing different rates of prohloraz (PRO), BAP, IBA and GA3, included macro and micro salts according to Murashige and Skoog (1962) (MS). PRO was applied at 3 concentrations (1,5 and 10 μM), either individually or combined with BAP (4.4 μM), IBA (5 μM) and GA3 (0.3 μM). Upon the second subculture, the parameters of shoot multiplication, multiplication index, and length and number of axial and lateral shoots were checked. Fresh and dry shoot weight was also evaluated. The highest multiplication index (...1:4.94) and fresh/dry shoot weight were observed on the medium containing PRO 5 μM, BAP 4.4 μM and GA3 0.3 μM. The fungicide, applied either individually or combined with IBA or GA3, had no effect on the multiplication of Gisela 6 sweet cherry rootstock regardless of the applied PRO concentration. Nevertheless, all studied concentrations of PRO combined with IBA affected rhizogenesis whereby the highest rooting rate was obtained on the medium containing PRO 10 μM (83.33%). The obtained results suggest that PRO intensifies the effect of exogenous added BAP on the multiplication of Gisela 6 vegetative sweet cherry rootstock, whereas when combined with IBA, PRO has been re ported to affect rhizogenesis.
