AB  - Nova sorta višnje ‘Sofija’, priznata je rešenjem Ministarstva polјoprivrede i zaštite životne sredine RS br. 320-04-00566/2011-11 od 19. januara 2015. godine. Stvorena planskom hibridizacijom iz ukrštanja ‘Čačanski rubin’ × ‘Heimanns Konserven Weichsel’, ova sorta je izdvojena u grupu od deset elitnih hibrida pod oznakom XII/57, iz populacije od preko 3.000 hibridnih sejanaca. Stablo sorte ‘Sofija’ je umerene bujnosti, sa karakterističnim povijenim granama, srednje ranog cvetanja. Plodovi su veoma krupni (7‒8 g), zaoblјenog oblika, slabo izraženog šava. Pokožica ploda je rubin crvene boje, glatka i sjajna, a mezokarp crvene boje, slatko-nakiselog ukusa i odličnog kvaliteta. Sok je crvene boje. Koštica je srednje krupnoće, a petelјka izrazito duga, tanka i lako se odvaja od ploda, bez curenja soka. ‘Sofija’ se odlikuje visokim stepenom polјske otpornosti na Blumeriella jaapii (Rehm) v. Arx. Ono što je od posebnog značaja za ovu sortu jeste njeno veoma interesantno vreme sazrevanja (početak druge dekade juna u uslovima zapadne Srbije). Imajući u vidu činjenicu da aktuelni sortiment višnje nije previše dinamičan, kao i da je vreme sazrevanja najvećeg broja komercijalno značajnih krupnoplodih sorti vezano za treću dekadu juna, plodovi sorte ‘Sofija’ zbog ranijeg vremena sazrevanja, uz istovremeno izvanredan kvalitet ploda i visoku rodnost, ostvaruju visoku cenu na tržištu. Od 2019. godine, sorta ‘Sofija’ je standardni deo asortimana u proizvodnji sadnog materijala višnje u Republici Srbiji, a od 2023. godine i van granica naše zemlje.
AB  - The new sour cherry cultivar ‘Sofija’ was named and released by the Ministry of Agriculture, Forestry, and Water Management of the Republic of Serbia (confirmation No. 320-04-00566/2011-11; January 11th, 2015). Developed by planned hybridization from the crossing of ‘Čačanski rubin’ × ‘Heimanns Konserven Weichsel’, this cultivar was selected into the group of ten elite hybrids (under the designation XII/57), from a population of over 3,000 hybrids. The tree of the ‘Sofia’ cultivar is moderately vigorous, with characteristic bent branches, of medium-early flowering. The fruits are very large (7‒8 g), rounded, with a weak seam. The skin of the fruit is ruby red in color, smooth and shiny, and the mesocarp is red, with a sweet-sour taste and excellent quality. The juice is red. The stone is of medium size, and the stem is very long, thin and easily separated from the fruit, without leaking juice. ‘Sofia’ is characterized by a high degree of field resistance to Blumeriella jaapii (Rehm) v. Arch. What is of special importance for this cultivar is its very interesting ripening time (the beginning of the second decade of June in the conditions of western Serbia). Bearing in mind the fact that the current sour cherry assortment is not too dynamic, as well as that the ripening time of the largest number of commercially significant large-fruited cultivars is related to the third decade of June, the fruits of the ‘Sofia’ cultivar, due to their earlier ripening time, with at the same time outstanding fruit quality and high fertility, achieve high price on the market. Since 2019, the variety ‘Sofija’ is a standard part of the assortment in the production of cherry planting material in the Republic of Serbia, and from 2023 also outside the borders of our country.

AB  - According to the average annual production, sour cherries take the third place in Serbian fruit production. In the assortment, the most represented is the autochthonous variety ‘Oblačinska’, which is characterized by a very small fruit. In recent decades, large-fruited varieties bred at the Fruit Research Institute in Čačak, have become more widespread in sour cherry orchards. Some of the newly bred varieties, similar to ‘Oblačinska’, are characterized by combined properties and can be used as fresh fruit and/or for various types of processing. Sour cherries are mainly used in Serbia as raw material for obtaining numerous products. Frozen cherries dominate, but juices, sour cherry in brine, dried fruits, candied fruits, compote, wine, vinegar, liqueurs are also produced, and in recent years, sour cherry spirit is gaining interest. The paper presents the results of two-year examinations of the suitability of two varieties of sour cherries with combined properties - ‘Šumadinka’ (‘Köröser Weichsel’ × ‘Heimanns Konservenweichsel’) and ‘Sofija’ (‘Čačanski rubin’ × ‘Heimanns Konservenweichsel’), bred in Čačak, and standard variety ‘Heimanns Konservenweichsel’, for the production of spirits. The sour cherry spirits were produced from spontaneously fermented mashes of sour cherry fruits with stones, followed by double distillation in alembic of traditional design. The obtained monovarietal sour cherry spirits differed significantly in spirit yields and contents of the ten major volatile components. Based on the results of sensory analysis in both years, all obtained monovarietal sour cherry spirits can be classified as high-quality spirit drinks.

An overview of S-genotype diversity in sweet cherry landraces grown in the central region of the Republic of Serbia

AB  - Identification of the S-genotypes in landraces is a crucial step in the molecular characterization of Serbian autochthonous sweet cherry germplasm. It is also of enormous significance for breeders and growers, as this fruit species exhibits a gametophytic self-incompatibility, controlled by the multi-allelic two genes of the S-locus. The aim of this study was to summarize known information and reveal new data on the S-alleles in 23 sweet cherry landraces originating in the Republic of Serbia. The use of polymerase chain reaction (PCR) with consensus primers for the second intron of S-RNase, primers specific for S-RNase and certain SFB alleles, along with DNA fragment analysis using fluorescently labelled forward primers to amplify both the S-RNase first intron and the SFB intron, revealed 10 alleles (S1 to S6, S9, S12, S13 and S22) that generated the following 13 S-genotypes: S1S2 (one landrace), S1S4 (one landrace), S1S5 (one landrace), S2S3 (four landraces), S3S4 (two landraces), S3S5 (two landraces), S3S6 (three landraces), S3S9 (two landraces), S3S12 (two landraces), S4S5 (one landrace), S4S13 (one landrace), S5S22 (one landrace) and S6S9 (two landraces). The most frequent S-allele and S-genotype in this sweet cherry material were S3 and S2S3, with occurrence frequencies of 32.6% and 17.4%, respectively. Based on the obtained results, the sweet cherry landraces were assigned to 12 incompatibility groups and one group of universal donors (ʻ0ʼ). These results provide important information about their cross-compatibility and the diversity of the S-locus in Serbian sweet cherry germplasm.

Stoga je ekspresija gena S-lokusa od esencijalnog značaja za regulisanje genetičke kompatibilnosti sorti trešnje, koje su međusobno inkompatibilne ukoliko su istog S-haplotipa. Za trešnju je karakteristično da postoji određeni broj samooplodnih sorti koje mogu zametati plodove pri oprašivanju sopstvenim polenom i istovremeno, zahvaljujući mutaciji u SFB genu, predstavljaju univerzalne oprašivače za ostale sorte. Nа оsnоvu S-gеnоtipа, sоrtе trеšnjе su klasifikovane u do sada poznate 63 grupe inkompatibilnosti (оznаčеnе оd I dо LXIII), kao i grupu sаmооplоdnih sоrti i grupu „0”, kојој pripаdајu sоrtе rеtkоg S-gеnоtipа kојi im оmоgućаvа dа budu pоtеnciјаlnо dоbri оprаšivаči sоrtаmа drugih grupа. Identifikacija S-genotipova kod autohtonih genotipova trešnje je ključan korak u molekularnoj karakterizaciji materijala kolekcionisanog na prostoru Republike Srbije, i od ogromnog je značaja kako za oplemenjivače, tako i za proizvođače zainteresovane za proizvodnju kvalitetnih genotipova adaptiranih na određene agro-ekološke uslove. Cilj ovog istraživanja bio je da sažme poznate informacije i otkrije nove podatke o S-alelima kod 23 autohtona, odnosno gеnоtipa trešnje nеpоznаtоg pоrеklа gajenih na različitim lokalitetima centralne Srbije. Genomska DNK ispitivanih genotipova trešnje je izolovana iz uzoraka mladog lista, primenom modifikovane CTAB mini prep metode. Upotreba lančane reakcije polimeraze (PCR) sa konsenzus prajmerima za drugi intron S-RNaze, prajmerima specifičnim za S-RNazu i određene alele SFB gena, zajedno sa DNK fragment analizom sa fluorescentno obeleženim prajmerima za amplifikaciju prvog introna S-RNaze i SFB introna, otkrila je 10 alela (S1 do S6, S9, S12, S13 i S22) koji su generisali 13 S-genotipova: S1S2 (jedan genotip), S1S4 (jedan genotip), S1S5 (jedan genotip), S2S3 (četiri genotipa), S3S4 (dva genotipa), S3S5 (dva genotipa), S3S6 (tri genotipa), S3S9 (dva genotipa), S3S12 (dva genotipa), S4S5 (jedan genotip), S4S13 (jedan genotip), S5S22 (jedan genotip) i S6S9 (dva genotipa). U ispitivanom autohtonom materijalu ustanovljeno je da je S3 najzastupljeniji alel, sa frekvencijom od 32,6%, dok je S2S3 najčešći S-genotip, sa učestalošću od 17,4%. Na osnovu dobijenih rezultata, autohtoni genotipovi trešnje su svrstani u 12 grupa inkompatibilnosti i grupu univerzalnih donora („0”). Rezultati ovog rada pružaju važne informacije o njihovoj međusobnoj kompatibilnosti i diverzitetu S-lokusa kod genotipova trešnje decenijama gajenih na prostoru naše zemlje.

abstract = "Identification of the S-genotypes in landraces is a crucial step in the molecular characterization of Serbian autochthonous sweet cherry germplasm. It is also of enormous significance for breeders and growers, as this fruit species exhibits a gametophytic self-incompatibility, controlled by the multi-allelic two genes of the S-locus. The aim of this study was to summarize known information and reveal new data on the S-alleles in 23 sweet cherry landraces originating in the Republic of Serbia. The use of polymerase chain reaction (PCR) with consensus primers for the second intron of S-RNase, primers specific for S-RNase and certain SFB alleles, along with DNA fragment analysis using fluorescently labelled forward primers to amplify both the S-RNase first intron and the SFB intron, revealed 10 alleles (S1 to S6, S9, S12, S13 and S22) that generated the following 13 S-genotypes: S1S2 (one landrace), S1S4 (one landrace), S1S5 (one landrace), S2S3 (four landraces), S3S4 (two landraces), S3S5 (two landraces), S3S6 (three landraces), S3S9 (two landraces), S3S12 (two landraces), S4S5 (one landrace), S4S13 (one landrace), S5S22 (one landrace) and S6S9 (two landraces). The most frequent S-allele and S-genotype in this sweet cherry material were S3 and S2S3, with occurrence frequencies of 32.6% and 17.4%, respectively. Based on the obtained results, the sweet cherry landraces were assigned to 12 incompatibility groups and one group of universal donors (ʻ0ʼ). These results provide important information about their cross-compatibility and the diversity of the S-locus in Serbian sweet cherry germplasm., Trešnja (Prunus avium L.) je samobesplodna vrsta voćaka, čija je auto-inkompatibilnost gametofitnog tipa, regulisana ekspresijom dva gena S-lokusa odgovornih za sintezu proteina S-RNaze i SFB, odnosno ženske i muške komponente specifičnosti prepoznavanja sopstvenog i nesopstvenog polena. Stoga je ekspresija gena S-lokusa od esencijalnog značaja za regulisanje genetičke kompatibilnosti sorti trešnje, koje su međusobno inkompatibilne ukoliko su istog S-haplotipa. Za trešnju je karakteristično da postoji određeni broj samooplodnih sorti koje mogu zametati plodove pri oprašivanju sopstvenim polenom i istovremeno, zahvaljujući mutaciji u SFB genu, predstavljaju univerzalne oprašivače za ostale sorte. Biological and pomological characteristics of autochthonous plum cultivars collected in western Serbia

AB  - During the 2020 and 2021, the flowering phenophase and the most important characteristics of trees and fruits from fourteen in situ collected autochthonous plum genotypes grown in the area of western Serbia were investigated. The obtained results revealed that none of the collected genotypes showed low vigour, while the suckering tendency varied from low to extremely high, and yielding efficiency ranged from intermediate to good. All investigated genotypes were characterized with long blooming period with genotypes ʽŠ-GMNe/1ʼ and ʽŠ-UžK/5ʼ standing out for their very late onset of flowering. The tested genotypes had oval to elliptical fruit shape, small stone and medium flesh percentage. In terms of fruit size, the most of genotypes belonged to the small fruit group, a few were categorized as medium-sized fruit and only one genotype (ʽŠ-KrGP/2ʼ) fell into the large fruit category. Regarding the content of soluble solids, all tested genotypes showed good fruit quality. However, the genotype ʽŠ-ČaB/1ʼ stood out for having the highest levels of soluble solids, total and invert sugars indicating its distinct chemical composition.
AB  - Republika Srbija obiluje autohtonim sortama i populacijama domaće šljive koje su nastale kao rezultat dugotrajne selekcije od strane čoveka, kao i od strane edafskih, klimatskih i geomorfoloških uslova karakterističnih za područje u kom su nastale. One predstavljaju ogromno nacionalno bogatstvo i prirodni resurs koji nije u dovoljnoj meri istražen, a samim tim ni upotrebljen u adekvatne svrhe. Čak šta više, sve je prisutniji trend gubitka genetičkih resursa uzrokovan prenamenom zemljišta i uvođenjem u proizvodnju novijih sorti visoke rodnosti. Rad na kolekcionisanju i proučavanju autohtonih genotipova šljive sa ciljem njihovog korišćenja u oplemenjivačkim programima i uvođenja u proizvodnju sprovodi se u Institutu za voćarstvo, Čačak od njegovog osnivanja do danas. U ovom radu su prikazani rezultati proučavanja najznačajnijih osobina stabla i ploda 14 autohtonih genotipova šljive nepoznatog porekla koji su, kao pojedinačna stabla gajena na sopstvenom korenu, in situ kolekcionisani na teritoriji Zapadne Srbije. Nazivi kolekcionisanih genotipova su formirani na osnovu geografskih odrednica (grad, opština i selo) i brojeva koji su dodeljeni prema redosledu sazrevanja. Najznačajnije osobine stabla (bujnost, tendencija ka davanju izdanaka i koeficijent rodnosti ispitani su prema metodologiji navedenoj u međunarodno priznatom deskriptoru (Descriptor List for Plum and Allied Species, IBPGR), dok su fenološke (početak i kraj cvetanja) i pomološke osobine (masa ploda i koštice, randman ploda, dimenzije i indeks oblika ploda, sadržaj rastvorljive suve materije, ukupnih i invertnih šećera, sadržaj saharoze, ukupne kiseline i pH soka ploda) ispitane korišćenjem standardnih metoda. Dobijeni rezultati su pokazali da u kolekcionisanom materijalu nema genotipova koji se odlikuju umanjenom bujnošću, dok je sklonost ka davanju izdanaka varirala od male do veoma velike. Proučavani genotipovi su tokom perioda ispitivanja (2020/21) postigli osrednju do dobru rodnost. Svi proučavani genotipovi šljive su se odlikovali dugim cvetanjem, a kod genotipova ʽŠ-GMNe/1ʼ i ʽŠ-UžK/5ʼ je uočen veoma kasni početak cvetanja. Većina kolekcionisanih genotipova se odlikovala eliptičnim oblikom ploda, sitnom košticom i srednjim randamanom ploda. Grupa genotipova sitnog ploda je bila najbrojnija, dok se nekoliko genotipova odlikovalo srednje krupnim plodom, a samo jedan genotip (ʽŠ-KrGP/2ʼ) krupnim plodom. Rezultati koji se odnose na sadržaj rastvorljive suve materije u plodu ukazuju na to da su se svi kolekcionisani genotipovi odlikovali dobrim kvalitetom ploda. U ovom pogledu prednjači genotip ʽŠ-ČaB/1ʼ u čijim plodovima je utvrđen najviši sadržaj rastvorljive suve materije, ukupnih i invertnih šećera.

Performance in vitro of fresh and stored plum (Prunus domestica L.) pollen

AB  - In this study performance in vitro of fresh and pollen stored for three months at different constant temperatures were investigated in three plum cultivars (ʻValerijaʼ, ʻČačanska Lepoticaʼ, ʻValjevkaʼ). Pollen were collected from the flowers in late balloon phase and after drying stored for three months at four different constant temperatures (+4 °C, ‒20 °C, ‒80 °C, ‒196 °C). Pollen tube length was examined as well as the nuclear presence in pollen tube and its migration. In vitro pollen tube length was tested after a 24-hour germination period after which the pollen tube was stained with Hoechst 33258 to observe the nuclear presence within it. The longest average pollen tube length in all tested cultivars were observed with fresh pollen. With the pollen stored at low temperatures the longer pollen tube was obtain compared to pollen stored at +4 °C. In all cultivars mature pollen grains were in binucleate stage contained vegetative nucleus (VN) and generative nucleus (GN). Although at each storage temperature a higher percentage of GN was achieved in pollen tubes compared to fresh ones, nevertheless, regular movement of nuclei, primarily VN than GN or sperm nucleus (SN), was found in the highest percentage in all tested cultivars.
AB  - U ovom radu su ispitane performanse in vitro svežeg i polena čuvanog na različitim konstantnim temperaturana tokom tri meseca kod tri sorte šljive (ʻValerijaʼ, ʻČačanska Lepoticaʼ, ʻValjevkaʼ). Polen je sakupljan iz cvetova koji su bili u kasnoj balon fazi i nakon sušenja čuvan na četiri konstantne temperature (+4 °C, ‒20 °C, ‒80 °C, ‒196 °C). Ispitana je dužina polenovih cevčica, prisustvo i migracija jedara u njima. Dužina polenovih cevčica je određivana nakon inkubacionog perioda od 24h a zatim bojenje sa Hoechst 33258 da bi se uočilo prisustvo jedara u njima. Prosečno najduže polenove cevčice kod svih ispitivanih sorti utvrđene su kod svežeg polena. Kod polena čuvanog na niskim temperaturama prosečna dužina polenovih cevčica je bila veća nego kod polena čuvanog na +4 °C. Polen svih sorti u zrelom stanju je dvojedaran, sadrži vegetativno i generativno jedro. Iako je kod svih sorti nakon perioda čuvanja polena, uočen veći procenat prisustva genetativnog jedra u polenovim cevčicama, raspored jedara je bio regularan, prvo vegetativno, potom generativno ili spermatično jedro.

abstract = "In this study performance in vitro of fresh and pollen stored for three months at different constant temperatures were investigated in three plum cultivars (ʻValerijaʼ, ʻČačanska Lepoticaʼ, ʻValjevkaʼ). Pollen were collected from the flowers in late balloon phase and after drying stored for three months at four different constant temperatures (+4 °C, ‒20 °C, ‒80 °C, ‒196 °C). Pollen tube length was examined as well as the nuclear presence in pollen tube and its migration. In vitro pollen tube length was tested after a 24-hour germination period after which the pollen tube was stained with Hoechst 33258 to observe the nuclear presence within it. The longest average pollen tube length in all tested cultivars were observed with fresh pollen. With the pollen stored at low temperatures the longer pollen tube was obtain compared to pollen stored at +4 °C. In all cultivars mature pollen grains were in binucleate stage contained vegetative nucleus (VN) and generative nucleus (GN). Although at each storage temperature a higher percentage of GN was achieved in pollen tubes compared to fresh ones, nevertheless, regular movement of nuclei, primarily VN than GN or sperm nucleus (SN), was found in the highest percentage in all tested cultivars., U ovom radu su ispitane performanse in vitro svežeg i polena čuvanog na različitim konstantnim temperaturana tokom tri meseca kod tri sorte šljive (ʻValerijaʼ, ʻČačanska Lepoticaʼ, ʻValjevkaʼ). Polen je sakupljan iz cvetova koji su bili u kasnoj balon fazi i nakon sušenja čuvan na četiri konstantne temperature (+4 °C, ‒20 °C, ‒80 °C, ‒196 °C). Ispitana je dužina polenovih cevčica, prisustvo i migracija jedara u njima. Dužina polenovih cevčica je određivana nakon inkubacionog perioda od 24h a zatim bojenje sa Hoechst 33258 da bi se uočilo prisustvo jedara u njima. Prosečno najduže polenove cevčice kod svih ispitivanih sorti utvrđene su kod svežeg polena. Kod polena čuvanog na niskim temperaturama prosečna dužina polenovih cevčica je bila veća nego kod polena čuvanog na +4 °C. Polen svih sorti u zrelom stanju je dvojedaran, sadrži vegetativno i generativno jedro. Iako je kod svih sorti nakon perioda čuvanja polena, uočen veći procenat prisustva genetativnog jedra u polenovim cevčicama, raspored jedara je bio regularan, prvo vegetativno, potom generativno ili spermatično jedro.",

AB  - Decades-long cherry breeding work at Fruit Research Institute, Čačak (FRI), implies the use as a main method planned hybridization within P. avium and P. cerasus. Combined with the selection from the natural populations, the method resulted in realization of two sweet cherry ('Asenova Rana' and 'Čarna') and five sour cherry cultivars ('Čačanski Rubin', 'Šumadinka', 'Sofija', 'Nevena' and 'Iskra'), three hybrids in the procedure of recognition, and numerous being intensively studied. Current sour cherry breeding work is based on the use of domestic genotypes, well adapted to the environmental conditions of the area (either obtained by planned hybridization or indigenous), and foreign genotypes – tolerant to diseases, with high cropping potential and good fruit quality. Different aspects of reproductive biology in cherries – flowering phenology, pollen tube growth in vitro and in vivo (pollen tube growth dynamics, pollen-pistil interactions in the style and ovary), cytoembryology (ovule development stage, viability of ovule and embryo sac, early embryogenesis), are among the main research activities at FRI. Within the past decade, the research on gametofitic self-incompatibility (GSI) in cherries has been advanced by identification of S-haplotypes in cultivars, landraces and hybrids using consensus, specific and dCAPS primers to reveal S-RNase and SFB functional/non-functional variants. Besides the GSI, other possible types of pollen-pistil interactions have also been considered, primarily in the context of temperature conditions during the flowering. Nowadays, within the CherrySeRB project whose FRI is the leading institution, characterization of the indigenous genotype potential for desirable reproductive properties and defining ʻgood reproductive behaviour cherry modelʼ that can face warmer temperature conditions during the flowering are in focus. The main CherrySeRB idea is connection of production challenges arising from climate change and the richness of Serbian and Balkan cherry germplasm that is still unused enough, which represents a novelty in breeding approach, applicable to other fruit species.

AB  - In sour cherry (Prunus cerasus L.), as in other species of the Prunus genus, the phenomenon of gametophytic self-incompatibility occurs. Basically similar to another Rosaceae species, sour cherry GSI is quite complex due to its tetraploid nature and the presence of pollen-part and stylar-part mutants that cause genetic changes at the S-locus. The phenomenon implies self-incompatible, partially self-compatible and self-compatible reproductive behavior of cultivars, as well as requirement for pollenisers in commercial orchards. Self-(in)compatibility status of sour cherry cultivars/selections realized at Fruit Research Institute, Čačak [‘Šumadinka’ (‘Köröser Weichsel’ × ‘Heimanns Konserven Weichsel’), ‘Sofija’ (‘Čačanski Rubin’ × ‘Heimanns Konserven Weichsel’), ‘Nevena’ (‘Köröser Weichsel’ × ‘Heimanns Konserven Weichsel’) and ‘GV-6’ (selected from natural population in the Čačak vicinity)] was examined over two years, and compared to standard ‘Heimanns Konserven Weichsel’. Self-pollination experiment in the field was conducted, as well as monitoring the parameters of pollen tubes growth efficacy in vivo by fluorescent microscopy (pollen tubes number and kinetics, fertilization percentage and presence of incompatible pollen tubes), and fruit set recording. In the pistils of ‘Nevena’, pollen tubes ended their growth predominantly in the upper part of the style, with sporadically penetrated tubes in the middle style. Pollen tubes kinetics was more efficient in the styles of other genotypes ‒ they were in the micropyle already on the 3rd day, and in the nucellus of the ovary on the 6th day after pollination (‘Šumadinka’, ‘Heimanns Konserven Weichsel’ and ‘Sofija’). According to fertilization percentage and fruit set, ‘Nevena’ could be undoubtedly classified within self-incompatible cultivars (both parameters were 0.00%, in both seasons), whereas ‘Šumadinka’ (17.92%; 16.31%, respectively) and ‘Heimanns Konserven Weichsel’ (18.75%; 17.50%, respectively) behaved as self-compatible. Showing relatively high values of fertilization percentage (21.05%; 23.18%, respectively), but lower values of fruit set (11.57%; 7.24%) after selfing, ‘Sofija’ and ‘GV-6’ could be classified as partially self-compatible genotypes.

Influence of temperature at the time of pollination on the effective pollination period and fruit set in plum

AB  - So far, the concept of effective pollination period (EPP) for several cultivars of plum, apricot, apple, pear, almond and others has been proven as suitable parameter to set up a relationship between the variation in the reproductive process and cropping behaviour. Among a number of autochthonous plum cultivar present in plum orchards in Serbia, ‘Crvena Ranka’ is the most important. It’s a self-sterile cultivar (characterized by the appearance of cytoplasmic male sterility), so the presence of an adequate pollenizer is needed. Fruit set and EPP in this plum cultivar were evaluated under natural field conditions in western Serbia during 2021 and 2022. Pollination of flowers with mix of dry pollen was subsequenly done during the first 10 days after full bloom. Initial and final fruit set was recorded four weeks after pollination and at the period of physiological maturity of the fruit, respectively. Results show that in both years number of initial fruit set increased up to five days after anthesis. The same was archived with the final fruit set in the first year while in the second, the higest number of fruit set was observed on the fourth day after anthesis (40.67% and 54.48%, resp.). In the first year, five days after anthesis a slightly deacrease in final fruit set was observed with the lowest, but still satisfactory values, on the 10th day after anthesis (11%). In the second year, decreasing in fruit set was more evident and was below 4% on the seventh day after anthesis. The reason for such significant decrease in fruit set was the spring frost occurred during the five days in a row. Taking into account clearly effects of weather conditions on fruit set, based on obtained results, it can be estimated that the EPP for plum cultivar ‘Crvena Ranka’  is 10 days.

Istraživanja reproduktivne biologije predstavnika roda Prunus, korišćenjem različitih metodologija (molekularnih, bioloških, histoloških), obezbedila su značajne informacije o biološkim događajima bitnim za zametanje plodova. Različiti aspekti koji se odnose na vreme cvetanja, potrebu za oprašivačima, muški i ženski gametofit, njihove performanse i interakcije (koje su u osnovi reproduktivnog uspeha) u različitim ekološkim kontekstima, pokazali su se kao veoma bitne teme od značaja za strategije oplemenjivanja i stvaranje novih sorti. Pokazalo se da su određene genetički determinisane reproduktivne osobine, kao što su kratko vreme cvetanja i životni vek ovula kao i, posledično, kratak efektivni polinacioni period, samobesplodnost, za modernu hortikulturnu praksu manje poželjne ili nepoželjne. Rezultati pokazuju da aktuelne klimatske promene imaju negativni uticaj na produkciju plodova u voćarstvu. Razmatrajući  sadašnju situaciju kao osnov za budući razvoj u oblasti oplemenjivanja i proizvodnje plodova koštičavih vrsta voćaka, neophodni su dalji integrativni pristupi. Novi ciljevi oplemenjivanja vrsta roda Prunus moraju objediniti adaptabilnost na temperaturne okolnosti koji proističu iz klimatskih promena, kvalitet ploda i otpornost na bolesti. U tom smislu, proučavanje fiziologije reproduktivnih procesa u narednim godinama će postajati sve značajnije, iz perspektive suočavanja sa promenama uslova životne sredine.
AB  - Considerable progress has been made in the breeding of stone fruits worldwide, in order to obtain cultivars that are adapted to different agro-ecological conditions, and with characteristics that ensure commercial effects and respond to consumer demands for healthy and quality fruits. Studies of the reproductive biology of Prunus species have combined different methodologies (molecular, biological, histological), providing valuable information on biological events essential for optimal fruit set. Different aspects related to blooming time, the needs for pollenisers, male and female gametophytes, their performances and interactions (that underlie reproductive success) under various ecological contexts, were shown to be important topics that contribute to the breeding strategies and developing of the new cultivars. Genetically determined reproductive traits such as short blooming time or ovule longevity and, consequently, insufficient effective pollination period, and self-incompatibility, have been shown to be less desirable or unsuitable from the aspect of modern horticultural practice. Nowadays, results also indicate that ongoing climate change has detrimental effects on fruit production. Considering different aspect of the current situation as a baseline for the future development of the stone fruits breeding and production, the future integrative approaches are necessary. The new breeding objectives of Prunus species must unify adaptability to temperature conditions arising from climate change, fruit quality and disease resistance. In this regard, the study of the physiology of reproductive processes will become increasingly important in future years from the perspective of coping with such environmental changes.

AB  - European plum is one of the most important temperate zone fruit species in the northern hemisphere. In the Republic of Serbia (RS), this species is beside apple and raspberry of great importance in terms of production, processing, consumption, export, employment of people and tradition. Therefore, constant improvement of plum production is necessary. The extension of the harvest season and the presence of fresh fruits on the market for a longer period is one of the significant aspects of that improvement. This is achieved by developing new cultivars with an earlier or later ripening time as well as cultivars that have better storage capability. Generally, cultivars with an earlier or later ripening time have a better price on the fresh fruit market. However, these cultivars, particularly of early ripening time have not been present to a significant extent in the orchards in the RS. Bearing this in mind, we investigate tree vigour (TCSA), productivity (yield per tree and yield efficiency), and the most important fruit physical (fruit and stone weight, flesh percentage, fruit dimensions, and sphericity) and chemical properties (contents of soluble solids, total and reducing sugars, saccharose and total acids, as well as ripening index and pH value) of three domestic ('Boranka', 'Čačanska Rana' and 'Valerija') and three introduced ('Katinka', 'Tegera' and 'Opal') early ripening cultivars which were harvested from the middle of the first to the end of the second decade of July. Obtained results showed that significant differences were found among cultivars for all examined characteristics. Overall, the assessed early-ripening cultivars could be grown to some extent in commercial orchards in the RS.

AB  - In this study, the most important characteristics of tree vigour and
productivity, as well as pomological traits (fruit morphometric and chemical
characteristics) of new late ripening cultivars ʻDivnaʼ and ʻPetraʼ released at Fruit
Research Institute, Čačak were evaluated and compared to standard cultivar
ʻStanleyʼ. Both new cultivars had slightly larger tree and yield in comparison to
ʻStanleyʼ, but smaller fruit in general. ʻPetraʼ had the best chemical properties of
fruit followed by ʻDivnaʼ. Both new cultivars could be very interesting for
growing in new orchards due to very late ripening time, good fruit quality and
high cropping potential and also could be used in further breeding activities.

AB  - Pollination and temperature are predominant factors affecting the progamic phase of sexual plant reproduction.
In light of climate change and the emergence of increasing temperatures, new knowledge is necessary to
encourage fruit breeding programmes to develop cultivars adapted to these changes, as well as to find the best
incompatibility relationships with the aim of achieving high fruit quality and yield. This research was conducted
to reveal the effect of pollination treatments and temperature on the progamic phase and fruit set in three European
plum (Prunus domestica L.) cultivars (‘Jojo’, ‘Hanita’ and ‘Katinka’) resistant/tolerant to Sharka virus
using fluorescent microscopy. Reproductive parameters (flowering overlapping, pollen germination, number of
pollen tubes in the upper part of the style and locule of the ovary, dynamics of pollen tubes growth and initial and
final fruit set) were investigated in vivo under different pollination treatments (cross, open and self-pollination).
The influence of different temperature regimes (20 ◦C, 23 ◦C and 26 ◦C) on the progamic phase was also observed
under cross and self-pollination. The pollination treatments significantly influenced the number of pollen tubes in
the style and ovary, as well as dynamics of pollen tubes in different regions of pistil. Pollen tubes showed better
growth in different crossing combinations compared to open and self-pollination. Higher temperatures accelerated
pollen tube growth and reduced the time they need to reach ovules. Generally, ‘Jojo’ appeared to be the
best pollenizer among the examined cultivars, while ‘Katinka’ exhibited the best response of pollen tube growth
to various temperature regimes. The differences observed between growth of the pollen tubes in each genotypetemperature
combination revealed that certain crossing combinations were better adapted to higher temperatures
so this can be taken into account as one of the adjustments to climate change. The ability of cultivars to
respond to high temperatures in the progamic phase can also be helpful for breeding programs in developing new
cultivars better adapted to different temperature conditions.

AB  - Within sweet cherry (Prunus avium L.), diversity is of the crucial importance in the context of adaptation to changing environmental conditions. The cultivars origin, their wider geographical distribution or spatial limitations in certain regions are related to their requirements for adequate conditions, which is especially visible through the reproductive process. This study was carried out to investigate pollen performance in vivo in the pistils of sweet cherry landrace of Balkan origin (‘Dolga Šiška’) over two-year period, in the conditions of prevailing Mediterranean climate of Ohrid Lake vicinity (Republic of North Macedonia). The pollenisers used are also of indigenous origin – ‘Ohridska Crna’ (Balkan region) and ‘Kordia’ (Czech Republic). Investigation was performed using the fluorescence microscopy method, on emasculated hand-pollinated flowers (cross-pollination) and non-emasculated open-pollinated flowers (open pollination), in three terms of pistil fixation (3rd, 6th and 10th day from the full flowering date). In general, ‘Ohridska Crna’ showed a tendency of inducing higher number of pollen tubes (22.14, 11.78 and 6.20 pollen tubes in the upper, middle and lower part of the style, respectively) in comparison with ‘Kordia’ (12.72, 6.62 and 2.92, respectively). This effect was particularly pronounced in the year with higher flowering temperature. On the other hand, pollen tube kinetics showed the opposite tendency, being better with ‘Kordia’ as polleniser in colder flowering season (7.69% of fertilization percentage and 3.60% fruit set, respectively), in comparison with ‘Ohridska Crna’ (penetration of pollen tubes into the nucellus and fruit set were not observed). Analysis of pollen performance in vivo parameters, pointed to the complex dependence on the polleniser genotype and temperature before and during the flowering, as well as the influence of pollination mode and pistilar genotype, which together contribute to the main factors’ interaction.

In situ characterization of plum landraces originated from the region of western Serbia

AB  - Eleven plum landraces (‘G-1’ to ‘G-11’) of unknown origin were sampled and analyzed as single trees on their own roots in orchards of the western Serbia region during 2020/21. The investigation included the most important agronomic traits such as harvest date, fruit and stone weight, fruit dimensions, surface area, soluble solids, and total acids as well as the ratio between them, total phenols and total anthocyanins, antioxidant activity, and field resistance to ‘Sharka’ disease (Plum pox virus). The collected material showed a wide range of variability in the studied properties. The investigated plum genotypes ripened between the 30th of July (‘G-1’) and the 15th of September (‘G-11’). The ‘G-3’ was characterized by the largest fruit (37.12 g), while the smallest fruit was found in ‘G-4’ and ‘G-5’ (11.76 g; 13.39 g). In terms of the fruit's chemical composition, the best results were obtained in ‘G-5’ and ‘G-11’ characterized by the highest content of soluble solids (20.45%; 21.10%) and the ratio between soluble solids and total acids (21.73; 30.14). The smallest content of soluble solids (14.25%) was found in ‘G-9’, while ‘G-3’ and ‘G-4’ had the smallest ratio between soluble solids and total acids (8.53; 9.59) due to the high value of total acids content (1.94%; 2.09%). The genotype ‘G-2’ was characterized by the highest content of total phenols (197.50 mg GAE 100 g-1 FW) and antioxidant activity (63.26%), while the value for total anthocyanins was highest in ‘G-5’ and ‘G-8’ (17.32 and 18.93 mg C3G 100 g-1 FW, respectively). The lowest level of bioactive compounds was found in ‘G-3’. Regarding the field resistance to ‘Sharka’ disease, the assessed genotypes can be divided into three groups, without symptoms (‘G-2’ to ‘G-4’, ‘G-6’ and ‘G-7’), with minor symptoms (‘G-1’, ‘G-8’ and ‘G-9’) and with moderate symptoms (‘G-5’, ‘G-10’ and ‘G-11’).

AB  - Sour cherry (Prunus cerasus L.) is one of the main perennial fruit crops in the Republic of Serbia (RS), whose average production of 117,595 tonnes (2012-2021) ranking it as the third most widely grown after plum and apple. As for the sour cherry assortment structure in the RS, clones of ‘Oblačinska’ are the most widespread (over 60%), while the remaining part comprises large-fruited foreign cultivars, as well as domestic ‘Šumadinka’ released within the breeding programme of the Fruit Research Institute, Čačak (FRI). As the tetraploid species, sour cherry includes both self-incompatible (SI) and self-compatible (SC) genotypes, which are characterised by more complex genetics of SI/SC. Therefore, developing of SC sour cherry cultivars is of high priority within FRI breeding work and worldwide. The aim of this study was to identify the S-haplotypes in two autochthonous (clones of ‘Oblačinska’ and ‘Cigančica’), four developed within FRI breeding programme (‘Iskra’, ‘Nevena’, ‘Sofija’ and ‘Šumadinka’) and one promising (‘GV-10’) sour cherry genotypes originated from the RS. The use of the polymerase chain reaction (PCR) method with three different consensus primer pairs for the first and the second introns of S-RNase, as well as primers specific for both S-RNase and SFB alleles, in combination with genetic tests to identify the pollen-part and stylar-part mutants (using derived cleaved amplified polymorphic sequence assay ‒ dCAPS), enabled identification of the S-haplotypes in the assessed sour cherry genotypes. In total, eight S-haplotypes (S1, S4, S6, S9, S13, S26, S35 and S36) or their non-functional variants were identified. In the investigated sour cherry genotypes, non-functional variant S36b2 was the most frequent (21.4%). The results generated in this study provide a valuable resource for design crosses in future breeding work which maximize the number of SC seedlings, and for orchard management in the efficient high-yielding fruit production.

AB  - Autochthonous genotypes of sour cherry (Prunus cerasus L.) are important source of genetic variability and represent valuable material for breeding work. In order to evaluate the main biological properties of ten sour cherry landraces (‘GV-6’, ‘GV-7’, ‘GV-8’, ‘GV-10’, ‘GV-11’, ‘GV-13’, ‘GV-14’, ‘GV-15’, ‘GV-16’, ‘GV-17’) from the West Serbia region, flowering and ripening time, fruit quality traits (morphometric, chemical and organoleptic), tree vigour, yield and field resistance to causal agents of diseases (cherry leaf spot and brown rot) were investigated. The highest average fruit weight was found in ‘GV-10’ and ‘GV-13’ (>7.5 g), whereas the highest soluble solids content was detected in ‘GV-7’ (17.30%). In terms of field resistance to pathogens, ‘GV-6’ and ‘GV-10’ showed the best performance. Owing to desirable biological and productive traits, some of the examined genotypes (‘GV-6’ and ‘GV-10’) have already been used as parents in the Fruit Research Institute, Čačak sour cherry breeding programme. Furthermore, ‘GV-6’ and ‘GV-10’ have potential for commercial production.
AB  - Autohtoni genotipovi različitih vrsta voćaka predstavljaju značajan izvor genetske varijabilnosti i vredan materijal za oplemenjivački rad. Institut za voćarstvo, Čačak ima dugogodišnju tradiciju u sakupljanju, evaluaciji i korišćenju autohtonog materijala različitih vrsta voćaka pa se u programu oplemenjivanja višnje, osim metode hibridizacije, koristi i selekcija iz prirodne populacije. Kako bi se ispitala pogodnost za komercijalno gajenje i/ili korišćenje u roditeljskim kombinacijama programa oplemenjivanja, ispitivane su biološke osobine deset genotipova višnje (GV-6, GV-7, GV-8, GV-10, GV-11, GV-13, GV-14, GV-15, GV-16, GV-17) izdvojenih na području Zapadne Srbije. Tokom dve godine (2020‡2021), ispitivani su vreme cvetanja i zrenja, morfometrijske, hemijske i organoleptičke osobine ploda, bujnost stabala, prinos i poljska otpornost na uzročnike bolesti (pegavost lista trešnje i mrka trulež). Najveću prosečnu masu ploda imali su genotipovi GV-10 i GV-13 (>7,5 g), dok je najveća vrednost sadržaja rastvorljive suve materije utvrđena kod genotipa GV-7 (17,30%). Genotipovi GV-6 i GV-10 pokazali su najveću poljsku otpornost na patogene. Zbog poželjnih bioloških i produktivnih osobina (krupan plod, duga peteljka, mali udeo koštice, visok sadržaj rastvorljive suve materije, izražena otpornost na patogene, visoka i redovna rodnost), genotipovi GV-6 i GV-10 imaju potencijal za komercijalnu proizvodnju. abstract = "Autochthonous genotypes of sour cherry (Prunus cerasus L.) are important source of genetic variability and represent valuable material for breeding work. In order to evaluate the main biological properties of ten sour cherry landraces (‘GV-6’, ‘GV-7’, ‘GV-8’, ‘GV-10’, ‘GV-11’, ‘GV-13’, ‘GV-14’, ‘GV-15’, ‘GV-16’, ‘GV-17’) from the West Serbia region, flowering and ripening time, fruit quality traits (morphometric, chemical and organoleptic), tree vigour, yield and field resistance to causal agents of diseases (cherry leaf spot and brown rot) were investigated. The highest average fruit weight was found in ‘GV-10’ and ‘GV-13’ (>7.5 g), whereas the highest soluble solids content was detected in ‘GV-7’ (17.30%). In terms of field resistance to pathogens, ‘GV-6’ and ‘GV-10’ showed the best performance. Owing to desirable biological and productive traits, some of the examined genotypes (‘GV-6’ and ‘GV-10’) have already been used as parents in the Fruit Research Institute, Čačak sour cherry breeding programme. Furthermore, ‘GV-6’ and ‘GV-10’ have potential for commercial production., Autohtoni genotipovi različitih vrsta voćaka predstavljaju značajan izvor genetske varijabilnosti i vredan materijal za oplemenjivački rad. Institut za voćarstvo, Čačak ima dugogodišnju tradiciju u sakupljanju, evaluaciji i korišćenju autohtonog materijala različitih vrsta voćaka pa se u programu oplemenjivanja višnje, osim metode hibridizacije, koristi i selekcija iz prirodne populacije. Kako bi se ispitala pogodnost za komercijalno gajenje i/ili korišćenje u roditeljskim kombinacijama programa oplemenjivanja, ispitivane su biološke osobine deset genotipova višnje (GV-6, GV-7, GV-8, GV-10, GV-11, GV-13, GV-14, GV-15, GV-16, GV-17) izdvojenih na području Zapadne Srbije. Tokom dve godine (2020‡2021), ispitivani su vreme cvetanja i zrenja, morfometrijske, hemijske i organoleptičke osobine ploda, bujnost stabala, prinos i poljska otpornost na uzročnike bolesti (pegavost lista trešnje i mrka trulež). Najveću prosečnu masu ploda imali su genotipovi GV-10 i GV-13 (>7,5 g), dok je najveća vrednost sadržaja rastvorljive suve materije utvrđena kod genotipa GV-7 (17,30%). Genotipovi GV-6 i GV-10 pokazali su najveću poljsku otpornost na patogene. Zbog poželjnih bioloških i produktivnih osobina (krupan plod, duga peteljka, mali udeo koštice, visok sadržaj rastvorljive suve materije, izražena otpornost na patogene, visoka i redovna rodnost), genotipovi GV-6 i GV-10 imaju potencijal za komercijalnu proizvodnju. AB  - Apple (Malus × domestica Borkh.) is one of the most important and widely cultivated deciduous fruit crop around the world, with an annual production of more than 93 million tonnes in 2021. This pome species, with average production of 468,258 tonnes (2017-2021) and together with plum, is crucial in the total fruit production of the Republic of Serbia. Although there are at least ten thousand reported apple cultivars in the world, as well as that new ones are realised every year within breeding programs, the number of cultivars is likely underestimated since many local landraces have not been documented. Identification of S-genotypes in autochthonous cultivars is an important step in their molecular characterization and also of enormous significance for breeders and growers, since apple exhibits a gametophytic self-incompatibility, controlled by the multi-allelic S-locus which prevents self-fertilisation. The aim of this work was to identify the S-genotypes in the following ten autochthonous apple cultivars grown in individual growers' orchards in the regions of central and southwestern Serbia: ‘Budimka’, ‘Džumurka’, ‘Ilinjača’ (Jezdina), ‘Ilinjača’ (Trnava), ‘Kolačara’, ‘Kraljica’, ‘Lepocvetka’, ‘Senabija’, ‘Šumatovka’ and ‘Vidovača’. The use of the polymerase chain reaction (PCR) method with consensus primers, that correspond to the previously identified C2 and C5 conserved regions, and allele-specific primers revealed eleven alleles: S1 to S5, S7, S20, S24, S26, S28, and S44. Among the assessed cultivars, the following ploidy levels at the S-locus were identified: four cultivars were diploids, three triploids and one was tetraploid; additionally, the S1Sx genotype was revealed in two cultivars. This study provides new information about cross-compatibility of cultivars and pollenizers, and might be used in parental choice within breeding programs, as well as in the choice of cultivars/pollenizers for the commercial orchards.

Quality of dried sour cherries from different Serbian cultivars

AB  - Dried fruit is classified as a nutritionally highly valued food due to its high content of phenolic compounds and antioxidant capacity. Furthermore, due to a high content of natural sugars giving the body the necessary energy, dried fruit is an essential part of a balanced meal. Therefore, the aim of our paper is to examine the quality of dried sour cherry from newly developed and indigenous cultivars originating from Serbia. Sour cherry cultivars and promising genotypes developed at Fruit Research Institute, Čačak ('Šumadinka', 'Sofija', 'Nevena' and 'GV-10') and autochthonous cultivars 'Oblačinska' and 'Feketićka' were used for testing. Drying of pitted fruit was performed in an experimental drier at an air temperature of 70 °C until reaching 75% of the total dry matter. Contents of total dry matter, sugars, total acids, as well as sugar/acid ratio and pH value were determined in fresh fruit. Large discrepancies manifested in the values of evaluated parameters point to distinct varietal specificities. Besides the aforementioned parameters, total phenols and antioxidant capacity, as well as sensory characteristics (appearance, flavour, aroma, and consistency) were analyzed in dried sour cherry fruits. Based on the results of sensory analyses of dried sour cherry fruits, the cultivar 'Feketićka' had the best, whereas cultivar 'Šumadinka' received the lowest grades, which is consistent with the sugar/acid ratio. On the other side, dried fruits of cultivar 'Feketićka' had the lowest values of contents of total phenolics and antioxidant capacity, whereas the highest values were found in cultivar 'Sofija'. Based on evaluated quality parameters, all tested cultivars have been found suitable for processing by drying. Sensory analysis revealed that the dried fruits of all tested sour cherries had supreme sensory quality.

AB  - The study was carried out to determine the effect of the site and temperature regime on the in vitro pollen performance of sweet cherries ʻLapins’, and ʻBurlat’, and sour cherry ʻŠumadinka’ in order to evaluate their ability as pollen donors for some Serbian cherry cultivars. For each cultivar pollen was collected from the two sites (Beograd and Čačak). In vitro pollen performance was conducted at 17°C and 20°C. Pollen germination was determined in a medium containing 0.7% agar-agar and 15% sucrose. For sweet cherries, increasing the temperature from 17°C to 20°C resulted in a significant decrease in pollen germination and pollen tube growth, while for ʻŠumadinka’ opposite result, but without a significant difference was observed. A significant influence of the site on pollen germination was established. Higher values were recorded for both sweet cherries in Čačak, while ʻŠumadinka’ showed a better response in Beograd. The shortest pollen tubes were observed in Burlat (370.6μm) while ʻŠumadinka’ had the longest ones (830.5μm). Site and temperatures affected pollen tube growth in vitro. Different germination percentages and pollen tube growth were found among both, different cultivars located in the same place and the same cultivars from two different locations. This leads to the different environmental conditions that influenced pollen formation in each location. More evaluation, including pollen performance in vivo and fruit settings in cross-pollinated combination with Serbian cherry cultivars is needed for the proper choice of pollen donors.

Phenological characteristics and fruit quality of introduced sweet cherry (Prunus avium L.) cultivars in agroecological conditions of Čačak

AB  - Phenological and pomological properties of nine introduced sweet cherry cultivars – ‘Burlat’, ‘Early Bigi’, ‘Early Lory’, ‘Early Star’, ‘Junska Rana’, ‘Newstar’, ‘Merchant’, ‘Sunburst’ and ‘Vega’, were studied in the vicinity of Čačak, during the two-year period (2018–2019). The investigation included morphometrical characteristics of fruit (fruit and stone weight; fruit dimensions – height, width and thickness; stalk length; fruit shape index; stone share), biochemical composition of fruit (soluble solids content; total sugars, inverted sugars and sucrose contents; total acids content; fruit juice pH, and sweetness index). The cultivars were characterized by medium-early to medium-late flowering time, and ripening span from 2nd to 4th ripening-week of cherry season. The highest value of fruit weight was found in ‘Sunburst’ (4th week of cherry ripening season) and ‘Early Star’ (2nd week of cherry ripening season). The chemical analyses also pointed to the high quality of fruits for these two cultivars, so they can be recommended for the commercial sweet cherry orchards in the Republic of Serbia, in order to improve assortment structure, and to extend harvesting season. ‘Merchant’ was also characterized with exceptional quality, in terms of biochemical composition of fruits. In relation to early ripening time (earlier than ‘Burlat’), cultivar ‘Early Bigi’ showed high values of quality parameters. Both ‘Merchant’ and ‘Early Bigi’ could be the part of sweet cherry assortment in a smaller extent, after ver-ification in production conditions, and in the areas where they give good results.
AB  - U cilju povećanja proizvodnje trešnje u Republici Srbiji, neophodno je uvođenje novih sorti koje u poređenju sa postojećim imaju atraktivniji izgled ploda i bolji kvalitet. Sorte treba da pripadaju različitim nedeljama sazrevanja trešnje, kako bi se tržištu obezbedili kvalitetni plodovi u što širem vremenskom periodu, a berba racionalno organizovala. U različitim programima oplemenjivanja u svetu posebna pažnja je posvećena dobijanju visokokvalitetnih ranih sorti trešnje koje mogu da konkurišu standardnoj sorti Burlat, odnosno da pokažu bolji kvalitet u pogledu krupnoće, čvrstine i otpornosti ploda na pucanje. Odlični oplemenjivački rezultati su postignuti sa sortama Early Lory i Early Bigi u Francuskoj, kao i sa sortom Early Star u Italiji. Fenološka i pomološka svojstva devet introdukovanih sorti trešnje (Burlat, Early Bigi, Early Lory, Early Star, Junska Rana, Newstar, Merchant, Sunburst i Vega), proučavana su tokom dvogodišnjeg perioda (2018–2019. godine) u okolini Čačka (lokalitet Jezdina). Ispitivanjima su obuhvaćene karakteristike fenofaza cvetanja i zrenja, morfometrijske karakteristike ploda (masa ploda i koštice; dimenzije ploda - visina,širina i debljina; dužina peteljke; indeks oblika ploda; udeo koštice u ukupnoj masi ploda), biohemijski sastav ploda (sadržaj rastvorljivih suvih materija; sadržaj ukupnih šećera, invertnih šećera i saharoze, sadržaj ukupnih kiselina, pH soka ploda i indeks slasti). Za ispitivane sorte je karakteristično srednje rano do srednje pozno vreme cvetanja, kao i vreme zrenja u drugoj (Early Bigi, Early Lory, Burlat, Early Star), trećoj (Junska Rana, Merchant, Newstar) i četvrtoj nedelji zrenja trešnje (Sunburst, Vega). Najbolje rezultate u pogledu krupnoće i biohemijskog sastava ploda pokazala je srednje-pozna samooplodna sorta Sunburst. Značajnu pažnju zaslužuje i Early Star, zbog ranijeg vremena sazrevanja i odličnih pomoloških karakteristika u odnosu na vreme zrenja. Označena kao sorta lokalnog značaja uglavnom zbog netšo manje čvrstine ploda, Merchant se odlikuje izuzetnim kvalitetom ploda i povoljnim vremenom sazrevanja, pa bi u manjem obimu mogao biti zastupljen u sortimentu trešnje, na lokalitetima gde daje dobre rezultate. Ispitivanja na različitim lokalitetima mogla bi da daju odgovor da li sorta Early Bigi, ranijeg sazrevanja u odnosu na sortu Burlat, može konkurisati ovoj sorti u pogledu kvaliteta ploda i drugih proizvodnih osobina. Unapređenje strukture sortimenta, uz korišćenje savremenog koncepta gajenja ove vrste, može značajno poboljšati kako ukupno stanje proizvodnje trešnje u Republici Srbiji, tako i voćarsku proizvodnju uopšte.

The cultivars were characterized by medium-early to medium-late flowering time, and ripening span from 2nd to 4th ripening-week of cherry season. The highest value of fruit weight was found in ‘Sunburst’ (4th week of cherry ripening season) and ‘Early Star’ (2nd week of cherry ripening season). The chemical analyses also pointed to the high quality of fruits for these two cultivars, so they can be recommended for the commercial sweet cherry orchards in the Republic of Serbia, in order to improve assortment structure, and to extend harvesting season. ‘Merchant’ was also characterized with exceptional quality, in terms of biochemical composition of fruits. In relation to early ripening time (earlier than ‘Burlat’), cultivar ‘Early Bigi’ showed high values of quality parameters. AB  - Fruit species of the Prunus genus are among the most represented worldwide. According to harvested
area and production volume, plum and sour cherry are beside apple and raspberry the most valuable
species in the Republic of Serbia. Also, peach, apricot and sweet cherry are of high importance.
Germplasms of all aforementioned Prunus species in the Republic of Serbia are very heterogeneous.
Numerous genotypes are characterized with high fruit quality and tolerance/resistance to causal agents of
the most important diseases and therefore, some of them such as plum ‘Crvena Ranka’, sour cherry
‘Obla~inska‘ and apricots ‘NS-4’ and ‘NS-6’ have a major impact on commercial production. On the other
hand, due to these properties, as well as others, a number of autochthonous genotypes have been used in
breeding work as one of the parents in planned hybridization. Selection of genotypes with better characteristics
have been carried out over a long time period in the Republic of Serbia and some of them, upon
evaluation under the recognition procedure, were named and released as new cultivars. Also, clonal selection
of the autochthonous genotypes and well-known landraces are a very useful tool for improvement of
assortment structure. Therefore, selected clones with better characteristics might be used in production and
further breeding work. Until these days, 11 apricot (‘^a~anska Pljosnata’, ‘^a~ansko Zlato’, ‘Aleksandar’,
‘Biljana’, ‘Vera’, ‘NS4’, ‘NS6’, ‘Novosadska Rodna’, ‘Novosadska Kasnocvetna’ ‘Buda’, ‘Ru`a’), two
peach (‘Ma{a’ and ‘Lela’), two sour cherry (‘Lenka’ and ‘Prima’) and one sweet cherry (‘Canetova’) genotypes
have been named and released as new cultivars. In addition, one plum, one sweet cherry and two sour
cherry genotypes are under the recognition procedure for new cultivars.
AB  - Vrste voćaka iz roda Prunus su me|u najznačajnijima
u svetu. Prema površini na kojoj se gaje i obimu proizvodnje,
šljiva i višnja su, uz jabuku i malinu, najznačajnije vrste voćaka u Republici Srbiji. Takođe, breskva,
kajsija i trešnja su od izuzetno velikog značaja.
Germplazma svih pomenutih vrsta u Republici Srbiji
je veoma raznolika i heterogena. Brojni genotipovi
različitih vrsta se između ostalog odlikuju izuzetnim
kvalitetom ploda i tolerantnošću/otpornošću na najznačajnije bolesti. Zbog svojih pozitivnih karakteristika
genotipovi Crvena ranka (šljiva), Oblačinska (višnja), NS-4 i NS-6 (kajsija) su od izuzetnog značaja za
komercijalnu proizvodnju. Takođe, ovi genotipovi,
kao i brojni drugi imaju veliki značaj i u oplemenjivanju
kao roditeljske sorte u planskoj hibridizaciji. Selekcija
genotipova boljih proizvodnih osobina iz prirodnih
populacija se u Republici Srbiji sprovodi duži
vremenski period i u taj proces su uključene sve relevantne
naučne institucije. U prethodnom periodu neki
genotipovi su izdvojeni i kroz postupak priznavanja
kod Komisije za priznavanje sorti pri Ministarstvu
poljoprivrede, šumarstva i vodoprivrede Republike Srbije
priznati su za nove sorte. Do sada je za nove sorte
priznato 11 genotipova kajsije (Čačanska Pljosnata,
Čačansko Zlato, Aleksandar, Biljana, Vera, NS-4, NS-
6, Novosadska Rodna, Novosadska Kasnocvetna, Buda
i Ruža), dva genotipa breskve, (Maša i Lela), dva
genotipa višnje (Lenka i Prima) i jedan genotip trešnje
(Canetova). Pored toga, u ovom momentu u proceduri
priznavanja za nove sorte nalaze se jedan genotip šljive
(Gardinovačka), jedan genotip trešnje (G-2) i dva
genotipa višnje (GV-6 i GV-10).

Preliminary results of in situ characterisation of autochthonous apple genotypes originated from the central and southwest Serbia region

AB  - The paper presents the main biological properties of 17 in situ old local apple cultivars and landraces grown in the regions of central and southwestern Serbia: ‘Ilinjača’ (Jezdina), ‘Kraljica’, ‘Vidovača’, ‘Ilinjača’ (Trnava), ‘J-GM/1’, ‘Prancija’, ‘Strekinja’, ‘Lepocvetka’, ‘J-LuN/1’, ‘Prancija Slatka’, ‘Džumurka’, ‘Slatka Kadumana’, ‘Lubenjaja’, ‘J-ČaO/1’, ‘Senabija’, ‘Budimka’ and ‘Kolačara’. The following characteristics of these genotypes were assessed: flowering phenophase, harvest date, pomological properties (physical and chemical, including bioactive compounds) and disease susceptibility to scab [Venturia inaequalis (Cooke) Wint.] and fireblight [Erwinia amylovora (Burnill)]. The average flowering time was during the third decade of April, whilst the predominant harvest date was from mid-September to mid-October. The highest fruit weight was determined in ‘Ilinjača’ (Trnava), while the lowest was in ‘Vidovača’ (262.99 g and 29.23 g, resp.). The best fruit quality among the assessed genotypes, measured by the fruit chemical composition, was found in ‘Senabija’ (soluble solids content ‒ 17.05%; total and invert sugars content ‒ 12.70% and 9.66%, resp.). The cultivar ‘Strekinja’ and landrace ‘J-ČaO/1’ were characterised by the highest content of total phenols and anthocyanins, respectively. In terms of fruit attractiveness, ‘Lepocvetka’, ‘J-ČaO/1’, ‘Senabija’ and ‘Budimka’ could be singled out. All of the studied genotypes showed resistance to fireblight and a relatively narrow range of susceptibility to scab under field conditions. Overall, Serbian autochthonous apple genotypes appear to carry useful characteristics that could be important for current/future breeding work and commercial growing suitable for fresh consumption, industrial and domestic processing.
AB  - Intenziviranjem procesa urbanizacije, odnosno prevashodno poljoprivredne proizvodnje kao njene funkcionalne komponente u skladu sa zahtevima globalnog svetskog tržišta, dolazi do neminovnog zapostavljanja autohtonog materijala, što za posledicu može imati drastičan ili nepovratan gubitak ovog dela dragocenog genofonda voćaka. Institut za voćarstvo, Čačak ima dugu tradiciju kolekcionisanja, proučavanja i usmerenog korišćenja divergentnosti autohtonog genofonda za oplemenjivačke programe, kao i programe direktnog uvođenja u proizvodnju. U cilju održavanja kontinuiteta proučavanja autohtonog materijala, u radu su prikazani rezultati ispitivanja najznačajnijih bioloških osobina 17 in situ genotipova jabuke gajenih na različitim lokalitetima teritorijalnih jedinica centralne i jugozapadne Srbije: Ilinjača (Jezdina), Kraljica, Vidovača, Ilinjača (Trnava), J-GM/1, Prancija, Strekinja, Lepocvetka, J-LuN/1, Prancija slatka, Džumurka, Slatka kadumana, Lubenjaja, J-ČaO/1, Senabija, Budimka i Kolačara. Primenom međunarodno priznatog deskriptora (Apple Descriptors, IBPGR) i tehničkog uputstva za jabuku (TG/14/9, UPOV), kao i korišćenjem standardnih laboratorijskih metoda ispitane su sledeće biološke osobine: fenofaza cvetanja, vreme berbe, pomološke osobine (fizičke i hemijske, uključujući i sadržaj bioaktivnih jedinjenja) i otpornost na prouzrokovače čađave pegavosti lista i krastavosti plodova [Venturia inaequalis (Cooke) Wint.], kao i bakteriozne plamenjače [Erwinia amylovora (Burnill)] u poljskim uslovima. Prosečno vreme cvetanja ispitivanih genotipova bilo je tokom treće dekade aprila, dok su plodovi sazrevali od 07. jula [Ilinjača (Jezdina)] do 18. oktobra (Kolačara). Najsitnijim plodom odlikovao se genotip Vidovača (masa ‒ 29,23 g, visina ‒ 43,81 mm i širina ‒ 41,04 mm), dok je najveću masu (262,99 g) i širinu ploda (87,39 mm) imao genotip Ilinjača (Trnava), a najveću visinu ploda genotip Lubenjaja (71,63 mm). U pogledu hemijskog sastava ploda, najviši sadržaj rastvorljivih suvih materija (RSM), ukupnih i invertnih šećera (17,05%, 12,70% i 9,66%, po redosledu) utvrđen je kod genotipa Senabija, dok je najviši sadržaj saharoze određen kod genotipa Budimka (3,66%). Najnižim sadržajem RSM odlikovali su se plodovi genotipa J-ČaO/1 (9,90%), dok su se plodovi genotipa Ilinjača (Jezdina) karakterisali najnižim sadržajem ukupnih šećera (5,28%) i saharoze (0,63%), uz istovremeno najviši sadržaj kiselina (1,52%) i najnižu pH vrednost soka ploda (2,94). Sadržaj ukupnih fenola u plodovima ispitivanih genotipova kretao se u intervalu od 117 do 337,50 mg galne kiseline na 100 g sveže mase ploda (Prancija i Strekinja, po redosledu). Plodovi genotipa Vidovača su se odlikovali najvišom antioksidativnom aktivnošću (89,84%), dok je najniža vrednost ovog parametra utvrđena u plodovima genotipa Džumurka (48,68%). Sadržaj ukupnih antocijana varirao je od 0,21 do 11,27 mg cijanidin-3-glukozida na 100 g sveže mase ploda (Prancija slatka i genotip crvenog mesa ploda J-ČaO/1, po redosledu). U pogledu atraktivnosti ploda, izdvajaju se genotipovi Lepocvetka, J-ČaO/1, Senabija i Budimka. Simptomi karakteristični za prouzrokovača bakteriozne plamenjače nisu uočeni u periodu proučavanja kod ispitivanih genotipova, dok su najviši stepen otpornosti na Venturia inaequalis u poljskim uslovima ispoljili genotipovi Kraljica, Strekinja, Džumurka, J-ČaO/1 i Senabija. U celini posmatrano, autohtoni genotipovi jabuke na prostoru Republike Srbije koji se odlikuju poželjnim biološkim osobinama obogatiće kolekciju početnog materijala za buduće oplemenjivačke programe, kao i za programe uvođenja u proizvodnju genotipova izvanrednih osobina namenjenih kako za svežu potrošnju, tako i za industrijsku i domaću preradu.

The following characteristics of these genotypes were assessed: flowering phenophase, harvest date, pomological properties (physical and chemical, including bioactive compounds) and disease susceptibility to scab [Venturia inaequalis (Cooke) Wint.] and fireblight [Erwinia amylovora (Burnill)]. The average flowering time was during the third decade of April, whilst the predominant harvest date was from mid-September to mid-October. The highest fruit weight was determined in ‘Ilinjača’ (Trnava), while the lowest was in ‘Vidovača’ (262.99 g and 29.23 g, resp.). The best fruit quality among the assessed genotypes, measured by the fruit chemical composition, was found in ‘Senabija’ (soluble solids content ‒ 17.05%; total and invert sugars content ‒ 12.70% and 9.66%, resp.). The cultivar ‘Strekinja’ and landrace ‘J-ČaO/1’ were characterised by the highest content of total phenols and anthocyanins, respectively. In terms of fruit attractiveness, ‘Lepocvetka’, ‘J-ČaO/1’, ‘Senabija’ and ‘Budimka’ could be singled out. All of the studied genotypes showed resistance to fireblight and a relatively narrow range of susceptibility to scab under field conditions. Overall, Serbian autochthonous apple genotypes appear to carry useful characteristics that could be important for current/future breeding work and commercial growing suitable for fresh consumption, industrial and domestic processing., Intenziviranjem procesa urbanizacije, odnosno prevashodno poljoprivredne proizvodnje kao njene funkcionalne komponente u skladu sa zahtevima globalnog svetskog tržišta, dolazi do neminovnog zapostavljanja autohtonog materijala, što za posledicu može imati drastičan ili nepovratan gubitak ovog dela dragocenog genofonda voćaka. Institut za voćarstvo, Čačak ima dugu tradiciju kolekcionisanja, proučavanja i usmerenog korišćenja divergentnosti autohtonog genofonda za oplemenjivačke programe, kao i programe direktnog uvođenja u proizvodnju. U cilju održavanja kontinuiteta proučavanja autohtonog materijala, u radu su prikazani rezultati ispitivanja najznačajnijih bioloških osobina 17 in situ genotipova jabuke gajenih na različitim lokalitetima teritorijalnih jedinica centralne i jugozapadne Srbije: Ilinjača (Jezdina), Kraljica, Vidovača, Ilinjača (Trnava), J-GM/1, Prancija, Strekinja, Lepocvetka, J-LuN/1, Prancija slatka, Džumurka, Slatka kadumana, Lubenjaja, J-ČaO/1, Senabija, Budimka i Kolačara. Primenom međunarodno priznatog deskriptora (Apple Descriptors, IBPGR) i tehničkog uputstva za jabuku (TG/14/9, UPOV), kao i korišćenjem standardnih laboratorijskih metoda ispitane su sledeće biološke osobine: fenofaza cvetanja, vreme berbe, pomološke osobine (fizičke i hemijske, uključujući i sadržaj bioaktivnih jedinjenja) i otpornost na prouzrokovače čađave pegavosti lista i krastavosti plodova [Venturia inaequalis (Cooke) Wint.], kao i bakteriozne plamenjače [Erwinia amylovora (Burnill)] u poljskim uslovima. Prosečno vreme cvetanja ispitivanih genotipova bilo je tokom treće dekade aprila, dok su plodovi sazrevali od 07. jula [Ilinjača (Jezdina)] do 18. oktobra (Kolačara). Predicting and optimising the quality of stored apples and maximising their shelf life are becoming a major challenge worldwide. Ethylene regulates a broad spectrum of developmental and physiological processes in apple (including fruit ripening) and being perceived through the family of receptors, among which Ethylene Receptor 1 (ETR1) is one of the Subfamily I receptors, encoded by ETR1 gene. This study aimed to identify the ETR1 alleles in nine Malus Miller species from the ex situ collection of the Fruit Research Institute, Čačak, Republic of Serbia. The ETR1 alleles were revealed by polymerase chain reaction (PCR) and additional restriction analysis of amplified PCR product (~5,000 bp) with RsaI, AluI and HinfI enzymes. Upon comparison of the observed polymorphisms, six alleles of ETR1 gene (a, b, c, d, f and g) were identified, among which f and g alleles were revealed in this study for the first time. Further characterisation of the novel alleles will be performed through cloning and sequencing of the corresponding DNA fragments. The polymorphisms observed upon PCR amplification and subsequent digestion with aforementioned restriction enzymes were generated in six allelic constitutions for ETR1 gene ‒ bb (one species), bd (one species), bf (one species), b,a/c (three species), ff (two species) and g,a/c/d/g (one species).
