AB  - The importance of conserving plant species, particularly those crucial for agriculture, has surged
to ensure sustainable utilization of biological resources and prevent further loss of plant diversity. Cryopreservation, the most advanced method for plant conservation, offers promise in this
regard. It can be employed for conservation per
se, or as a potential tool (cryotherapy) for pathogens elimination from infected plants, particularly viruses that threaten agricultural productivity. In the recent study, two vitrification-based
cryopreservation methods, D and V cryo-plate,
were evaluated for their efficacy in eradicating
plum pox virus (PPV) from autochthonous plum
cultivars ‘Crvena Ranka’ and ‘Belošljiva’, widely
present in the Balkan region. A total of 111 pool
samples of in vitro shoots of plums ‘Belošljiva’
and ‘Crvena Ranka’ were tested (65 and 46 samples, respectively). Each sample consisted of an
average of 10 plants, in total about 1100 plants.
Health status of in vitro shoots originated from
both control (non-frozen explants) and cryopreserved explants of infected plums were continuously tested during multiplication for the PPV
presence with conventional reverse transcription
polymerase chain reaction (RT-PCR). To evaluate
these results and obtain the highest efficiency of
detection, all samples were further tested using
Real-time PCR (qPCR). Using RT-PCR, the PPV was
detected in 67 out of 111 tested samples: in 39
samples of plum ‘Belošljiva’ and 28 of plum ‘Crvena Ranka’. Using a qPCR assay, PPV was confirmed
in 82 tested samples. Effective PPV elimination
from ‘Crvena Ranka’ was achieved through three
cryo-treatments, while neither method (D and
V-cryoplate) nor the applied treatments led to
PPV elimination in ‘Belošljiva’. The qPCR assay
demonstrated higher sensitivity compared to
conventional RT-PCR, highlighting its potential
utility in such assessments. Our findings supported the exceptional specificity and sensitivity of
the qPCR technique in detecting PPV in in vitro
shoots of the plums ‘Belošljiva’ and ‘Crvena Ranka’ post-cryotherapy.

Cryopreservation of highbush blueberry, strawberry, and saskatoon using V and D cryo‑plate methods and monitoring of multiplication ability of regenerated shoots

AB  - The purpose of this work was to apply V and D cryo-plate methods for cryopreservation of Vaccinium corymbosum ʻToroʼ, Fragaria × ananassa ʻCleryʼ, and Amelanchier alnifolia (Nutt.) M. Roem. and to monitor the multiplication capacity of shoots regenerated from cryopreserved explants. Shoot tips pre-cultured for 1 d at 23°C in the dark on medium containing 0.3 M sucrose were used as explants. Loading was performed in a solution containing 2 M glycerol and 0.8 M sucrose (30 min at room temperature). In the V cryo-plate, dehydration was carried out at room temperature (20 to 50 min) using the following plant vitrification solutions: original PVS2, 90% PVS2 solution, and PVS3. Regarding the D cryo-plate, dehydration was performed in closed glass containers over silica gel for 2, 2.5, or 3 h. In both protocols, rewarming was carried out in a 1.0 M sucrose solution (15 min at 25°C). Regenerated shoots were multiplied and multiplication parameters were monitored after the second subculture. Using the V cryo-plate method, the highest regrowth in highbush blueberry was obtained following 50-min treatment with all three VSs (61.7 to 80.9%). The D cryo-plate method was even more suitable with maximum regrowth of 89.4% achieved after 2.5 h of desiccation. For strawberry, 62.5% was the highest regrowth recorded using PVS3-based V cryo-plate method while 83.3% of regrowth was observed using D cryo-plate protocol. Regrowth of saskatoon reached a maximum of 50% after 50-min treatment with PVS3 while it did not exceed 40% in other treatments. By the second subculture, shoots regenerated from cryopreserved explants regained and even exceeded the multiplication capacity of shoots regenerated from non-cryopreserved explants. This study is the first to present the successful application of the V cryo-plate method in highbush blueberry, as well as the utilization of both V and D cryo-plate methods in saskatoon.

Performance in vitro of fresh and stored plum (Prunus domestica L.) pollen

AB  - In this study performance in vitro of fresh and pollen stored for three months at different constant temperatures were investigated in three plum cultivars (ʻValerijaʼ, ʻČačanska Lepoticaʼ, ʻValjevkaʼ). Pollen were collected from the flowers in late balloon phase and after drying stored for three months at four different constant temperatures (+4 °C, ‒20 °C, ‒80 °C, ‒196 °C). Pollen tube length was examined as well as the nuclear presence in pollen tube and its migration. In vitro pollen tube length was tested after a 24-hour germination period after which the pollen tube was stained with Hoechst 33258 to observe the nuclear presence within it. The longest average pollen tube length in all tested cultivars were observed with fresh pollen. With the pollen stored at low temperatures the longer pollen tube was obtain compared to pollen stored at +4 °C. In all cultivars mature pollen grains were in binucleate stage contained vegetative nucleus (VN) and generative nucleus (GN). Although at each storage temperature a higher percentage of GN was achieved in pollen tubes compared to fresh ones, nevertheless, regular movement of nuclei, primarily VN than GN or sperm nucleus (SN), was found in the highest percentage in all tested cultivars.
AB  - U ovom radu su ispitane performanse in vitro svežeg i polena čuvanog na različitim konstantnim temperaturana tokom tri meseca kod tri sorte šljive (ʻValerijaʼ, ʻČačanska Lepoticaʼ, ʻValjevkaʼ). Polen je sakupljan iz cvetova koji su bili u kasnoj balon fazi i nakon sušenja čuvan na četiri konstantne temperature (+4 °C, ‒20 °C, ‒80 °C, ‒196 °C). Ispitana je dužina polenovih cevčica, prisustvo i migracija jedara u njima. Dužina polenovih cevčica je određivana nakon inkubacionog perioda od 24h a zatim bojenje sa Hoechst 33258 da bi se uočilo prisustvo jedara u njima. Prosečno najduže polenove cevčice kod svih ispitivanih sorti utvrđene su kod svežeg polena. Kod polena čuvanog na niskim temperaturama prosečna dužina polenovih cevčica je bila veća nego kod polena čuvanog na +4 °C. Polen svih sorti u zrelom stanju je dvojedaran, sadrži vegetativno i generativno jedro. Iako je kod svih sorti nakon perioda čuvanja polena, uočen veći procenat prisustva genetativnog jedra u polenovim cevčicama, raspored jedara je bio regularan, prvo vegetativno, potom generativno ili spermatično jedro.

Evaluation of multiplication potential of cold stored shoot cultures of selected fruit genotypes

AB  - We presented the results about preserving in vitro selected fruit genotypes by cold storage strategy and the capacity for following shoot multiplication. The investigation included five fruit genotypes: plum ʻPožegača’, cherry rootstocks (ʻGisela 5’ and ʻTabel Edabriz’), raspberry ʻMeeker’, and blackberry ʻČačanska Bestrna’. The best results were achieved with shoots of the plum cultivar ʿPožegačaʾ which can be successfully stored at 5°C up to 10 months, without subculturing producing viable shoots. Furthermore, a significant increase in the multiplication index (13.0) of this genotype was noticed especially in shoots cultivated for 21 days under standard growing conditions, followed by storage at 5°C in total darkness for six months. ʻGisela 5’ and ʻTabel Edabriz’ cherry rootstocks were successfully preserved for three months in the same manner, and the plants (100%) well recovered. After being stored in cold conditions, the multiplication index of shoots in both rootstocks exhibited an increase ranging from 1.7 to 4.8 times compared to those grown under standard conditions. The viability rate of in vitro shoots of the raspberry cultivar ʻMeeker’ in cold conditions was also high (>67%). In this genotype, as well as in both cherry rootstocks, slow growth storage was accompanied by slow nitrogen uptake from the medium. In addition, the multiplication potential of encapsulated raspberry ʻMeeker’ and blackberry ʻČačanska Bestrna’ and cold stored (5°C, darkness) shoot tips was evaluated. Encapsulated shoot tips of ʻČačanska Bestrna’ (18.8%) and ʻMeeker’ (6.3%) survived the monthly cold storage period and exhibited regrowth. Shoots of selected genotypes were capable of multiplication in successive subcultures after cold storage, and often the multiplication indexes were higher compared to the control shoots that were consistently grown under standard conditions in the growth chamber. These results represent a significant contribution to the revitalization and further development of the national fruit gene bank.
AB  - U ovom radu su prikazani rezultati in vitro čuvanja kultura voćaka korišćenjem strategije hladnog skladištenja i sposobnosti izdanaka za multiplikaciju nakon konzervacije. U istraživanja je bilo uključeno pet genotipova različitih vrsta voćaka: sorta šljive Požegača, vegetativne podloge za trešnju i višnju Gisela 5 i Tabel Edabriz, sorta maline Meeker i sorta kupine Čačanska bestrna. Sorta šljive Požegača je uspešno čuvana do 10 meseci, dajući izdanke sa visokim indeksom multiplikacije koji je bio posebno izražen kod izdanaka uzgajanih 21 dan u standardnim uslovima, a zatim skladištenih na 5°C u potpunom mraku tokom šest meseci. AB  - In Serbia, the dominant methods of vegetative propagation of raspberries and blackberries are by root suckering (raspberry) and tip layering (blackberry), and they represent standard propagation techniques. However, due to the possible contamination of such planting material with viruses and phytopathogenic fungi, obtaining plants from tissue culture in vitro is becoming increasingly important. Therefore, the aim of this study was to investigate the fruit quality according to their chemical composition, emphasising the content of bioactive compounds in fruits of raspberry (Rubus idaeus L.) ‘Meeker’ and blackberry (Rubus fruticosus L.) ‘Čačanska Bestrna’ from plants
propagated in the field by standard technique (ST) and grown in vitro micropropagation tissue culture (TC). A three-year experiment included the determination of the main fruit quality parameters (soluble solids, sugars, and total acids) by standard methods. The identification and quantification of phenolic compounds (protocatechuic, 4-hydroxybenzoic, ellagic, gallic, p-coumaric, caffeic and ferulic acids, quercetin, cyanidin, and pelargonidin) were performed using high-performance liquid chromatography (HPLC). The obtained results showed that the origin of the planting material did not significantly affect the quality of the fruits in terms of primary and secondary metabolites, except for 4-hydroxybenzoic acid in blackberries. This means that both (ST and TC) propagation methods allow obtaining fruits of equally good quality. Therefore, when choosing the type of planting material, other factors such as health, yield, and cost of planting material should be taken into account.

Cryopreservation of indigenous plums and monitoring of multiplication and rooting capacity of shoots obtained from cryopreserved specimens

AB  - The objective of this study is to assess the suitability of vitrification cryo-plate (V cryo-plate) and dehydration cryo-plate (D cryo-plate) methods for the long-term conservation of eight autochthonous Prunus domestica L. genotypes originating from the Balkan Peninsula region. In vitro shoot tips were briefly pre-cultured for 1 day at 23 °C in the dark on a medium containing 0.3 M sucrose and then embedded in calcium alginate gel within the wells of the aluminum cryo-plates. In the V cryo-plate protocol, dehydration was carried out at room temperature using the following vitrification solutions: original plant vitrification solution 2 (PVS2) and 90% PVS2 solution (for 20 and 40 min) and plant vitrification solution 3 (PVS3) (for 60 and 80 min). In the D cryo-plate protocol, desiccation was performed for 2, 2.5, or 3 h over silica gel at 23 °C. The effect of different treatments was evaluated by monitoring the regrowth of both non-frozen and cryo-preserved explants. After cryo-preservation, five genotypes achieved regrowth rates over 40% in at least one of the applied protocols, while two genotypes showed regrowth rates of around 10%. A significant improvement in regrowth success for all genotypes using both cryo-plate methods was achieved by pre-culturing shoot tips for 7 days on a medium containing 0.5 M sucrose in complete darkness at 4 °C. Shoots regenerated from cryo-preserved explants were further monitored in vitro. By the third subculture, they had not only regained but had even exceeded the multiplication capacity (index of multiplication, length of axial, and lateral shoots) of shoots regenerated from dissection controls. abstract = "The objective of this study is to assess the suitability of vitrification cryo-plate (V cryo-plate) and dehydration cryo-plate (D cryo-plate) methods for the long-term conservation of eight autochthonous Prunus domestica L. genotypes originating from the Balkan Peninsula region. In vitro shoot tips were briefly pre-cultured for 1 day at 23 °C in the dark on a medium containing 0.3 M sucrose and then embedded in calcium alginate gel within the wells of the aluminum cryo-plates. In the V cryo-plate protocol, dehydration was carried out at room temperature using the following vitrification solutions: original plant vitrification solution 2 (PVS2) and 90% PVS2 solution (for 20 and 40 min) and plant vitrification solution 3 (PVS3) (for 60 and 80 min). In the D cryo-plate protocol, desiccation was performed for 2, 2.5, or 3 h over silica gel at 23 °C. The effect of different treatments was evaluated by monitoring the regrowth of both non-frozen and cryo-preserved explants. After cryo-preservation, five genotypes achieved regrowth rates over 40% in at least one of the applied protocols, while two genotypes showed regrowth rates of around 10%. A significant improvement in regrowth success for all genotypes using both cryo-plate methods was achieved by pre-culturing shoot tips for 7 days on a medium containing 0.5 M sucrose in complete darkness at 4 °C. Shoots regenerated from cryo-preserved explants were further monitored in vitro. By the third subculture, they had not only regained but had even exceeded the multiplication capacity (index of multiplication, length of axial, and lateral shoots) of shoots regenerated from dissection controls. Following multiplication, the cryo-preserved shoots were successfully rooted and rooting ability was assessed by monitoring the percentage of rooting, number and length of roots, and height of rooted plantlets.",

The most advanced method for plant conservation is cryopreservation. Cryotherapy that relies on cryopreservation procedure, alone or in combination with other techniques, can be used for pathogen elimination from infected plants. Viral diseases pose a significant threat to the agriculture, decreasing yields and crop quality. In this work, two cryotherapy methods (D and V cryo-plate) were evaluated for plum pox virus (PPV) eradication from autochthonous plums (Prunus domestica L.) 'Crvena Ranka' and 'Belosljiva', widely present under different local names on the Balkan peninsula. Nine different cryo-plate treatments were performed per genotype. Cryopreservation was effective in PPV elimination from variety 'Crvena Ranka' in three cryo-treatments. PPV elimination was, however, not achieved in variety 'Belosljiva' by any of the two methods (D and V-cryoplate) and treatments applied.

S-RNase genotyping of autochthonous apple cultivars grown in the region of central and southwestern Serbia

AB  - Apple (Malus × domestica Borkh.) is one of the most important and widely cultivated deciduous fruit crop around the world, with an annual production of more than 93 million tonnes in 2021. This pome species, with average production of 468,258 tonnes (2017-2021) and together with plum, is crucial in the total fruit production of the Republic of Serbia. Although there are at least ten thousand reported apple cultivars in the world, as well as that new ones are realised every year within breeding programs, the number of cultivars is likely underestimated since many local landraces have not been documented. Identification of S-genotypes in autochthonous cultivars is an important step in their molecular characterization and also of enormous significance for breeders and growers, since apple exhibits a gametophytic self-incompatibility, controlled by the multi-allelic S-locus which prevents self-fertilisation. The aim of this work was to identify the S-genotypes in the following ten autochthonous apple cultivars grown in individual growers' orchards in the regions of central and southwestern Serbia: ‘Budimka’, ‘Džumurka’, ‘Ilinjača’ (Jezdina), ‘Ilinjača’ (Trnava), ‘Kolačara’, ‘Kraljica’, ‘Lepocvetka’, ‘Senabija’, ‘Šumatovka’ and ‘Vidovača’. The use of the polymerase chain reaction (PCR) method with consensus primers, that correspond to the previously identified C2 and C5 conserved regions, and allele-specific primers revealed eleven alleles: S1 to S5, S7, S20, S24, S26, S28, and S44. Among the assessed cultivars, the following ploidy levels at the S-locus were identified: four cultivars were diploids, three triploids and one was tetraploid; additionally, the S1Sx genotype was revealed in two cultivars. This study provides new information about cross-compatibility of cultivars and pollenizers, and might be used in parental choice within breeding programs, as well as in the choice of cultivars/pollenizers for the commercial orchards.

Black currant propagation by softwood cuttings

AB  - Vegetative propagation of black currants by stem cuttings is mostly used by nurseries, as such method is quick and the newly propagated plants are genetically identical to parent plants. However, rooting in some genotypes is slow and results in a small number of rooted cuttings. In this study, softwood cuttings from three selected black currant cultivars (ʻBen Lomondʼ, ʻČačanska Crnaʼ and ʻTitaniaʼ) were subjected to different concentrations (250 ppm, 500 ppm and 1000 ppm) of rooting stimulators – indole-3-butyric acid (IBA) and α-naphthaleneacetic acid (NAA) before planting in rooting soil substrate. Cuttings without treatment were considered as control. Rooting performance of stated cultivars in tap water, without any previous treatment of cuttings, was examined as well. The following parameters were monitored: percentage of rooting, average number of roots per cutting, average root length and average height of newly formed shoots. All monitored parameters were significantly affected by the interaction cultivar × auxin treatment/ water rooting period. Rootingability of ʻBen Lomondʼ and ʻTitaniaʼ cuttings was improved after treatments with both IBA and NAA compared to non-treated cuttings. In both cultivars, IBA treatments gave better results than NAA. However, inhibition was observed in the case of ʻČačanska Crnaʼ cuttings treated with auxins compared with control (untreated) ones. Rooting ability of all cultivars, especially considering percentage of rooting and mean number of roots per cutting, was significantly improved by using a tap water as rooting medium without any previous treatment of cuttings.

AB  - Krioprezervacija predstavlja savremenu metodu za dugotrajno čuvanje biljnog materijala na ultra niskim temperaturama (tečni azot, -196 °C). Ova metoda je komplementarna konvencionalnim metodama čuvanja jer značajno snižava troškove i rizike vezane za održavanje biljnih kolekcija u poljskim uslovima (ex situ). Program primene ove strategije za dugotrajno očuvanje biljnih genetičkih resursa iniciran je i u Institutu za voćarstvo, Čaćak u okviru Odeljenja za fiziologiju voćaka pre više od jedne decenije. Istraživanja su fokusirana na optimizaciju protokola za različite in vitro tehnike krioprezervacije koje se baziraju na direktnom potapanju biljnog materijala u tečni azot i njihovu primenu u čuvanju genetičkih resursa kontinentalnih voćaka. Primenjene tehnike podrazumavaju primenu hemijske (vitrifikacija, „droplet“ vitrifikacija, „V cryo-plate“) ili fizičke dehidratacije (inkapsulacija-dehidracija, „D cryo-plate“) biljnog materijala pre potapanja u tečni azot. Tehnika inkapsulacije-dehidracije je uspešno primenjena u krioprezervaciji džanarike i sorte kupine Čaćanska bestrna. Ispitivanje primene tehnike vitrifikacija kod vrsta roda Prunus i Malus pokazalo je da se značajno povećanje kapaciteta regeneracije vrhova izdanaka podloge za trešnju Gisela 5 i sorte jabuke Gala Must posle otapanja iz tečnog azota može postići variranjem dužine dehidratacije i/ili vrste korišćenih vitrifikacionih rastvora. Optimizacija protokola za tehniku „droplet“ vitrifikacije postignuta je ispitivanjem uticaja različitih vitrifikacionih rastvora, dužine tretmana, temperature dehidratacije i dužine osmotske rehidratacije kod predstavnika rodova Prunus (autohtone sorte šljive Crvena Ranka, Sitnica i Požegača, džanarika, podloga za tešnju Gisela 5), Malus (jabuka Gala Must) i Rubus (kupina Čačanska bestrna). Poslednjih godina, inicirana su takođe i istraživanja primene novorazvijenih „V cryo-plate“ i „D cryoplate“ tehnika krioprezervacije. Ove tehnike omogućavaju prevazila`enje izvesnih ograničenja vezanih za nedovoljnu dehidrataciju biljnog tkiva, oštećenje i gubitak biljnog materijala tokom pripreme za krioprezervacione protokole, kao i manipulativnih problema u protokolima drugih tehnika vitrifikacije. Korišćenjem navedenih tehnika uspešno su krioprezervirani vrhovi izdanaka različitih genotipova autohtonih šjiva (Crvena Ranka, Požegača, Sitnica, Moravka, Belošljiva), džanarike, podloge za trešnju Gisela 5, sorte jagode Clery, borovnice Toro i mongolske borovnice (Amelanchier alnifolia) koji su gajeni u uslovima kulture in vitro. Dosadašnji rezultati postignuti u ovim istraživanjima predstavljaju značajan doprinos uspostavljanju optimizirane tehnologije dugotrajnog čuvanja biljnog materijala primenom različitih tehnika krioprezervacije. Ovaj pregled istraživanja daje dobru osnovu za dalju optimizaciju protokola koje će biti moguće direktno primeniti u budući program uspostavljanja in vitro banke gena (germplazme) voćnih biljnih vrsta koje se gaje u Srbiji., Cryopreservation represents contemporary method for long-term conservation of plant germplasm at ultralow temperature (liquid nitrogen, -196oC). Istraživanja su fokusirana na optimizaciju protokola za različite in vitro tehnike krioprezervacije koje se baziraju na direktnom potapanju biljnog materijala u tečni azot i njihovu primenu u čuvanju genetičkih resursa kontinentalnih voćaka. Primenjene tehnike podrazumavaju primenu hemijske (vitrifikacija, „droplet“ vitrifikacija, „V cryo-plate“) ili fizičke dehidratacije (inkapsulacija-dehidracija, „D cryo-plate“) biljnog materijala pre potapanja u tečni azot. Tehnika inkapsulacije-dehidracije je uspešno primenjena u krioprezervaciji džanarike i sorte kupine Čaćanska bestrna. Ispitivanje primene tehnike vitrifikacija kod vrsta roda Prunus i Malus pokazalo je da se značajno povećanje kapaciteta regeneracije vrhova izdanaka podloge za trešnju Gisela 5 i sorte jabuke Gala Must posle otapanja iz tečnog azota može postići variranjem dužine dehidratacije i/ili vrste korišćenih vitrifikacionih rastvora. Optimizacija protokola za tehniku „droplet“ vitrifikacije postignuta je ispitivanjem uticaja različitih vitrifikacionih rastvora, dužine tretmana, temperature dehidratacije i dužine osmotske rehidratacije kod predstavnika rodova Prunus (autohtone sorte šljive Crvena Ranka, Sitnica i Požegača, džanarika, podloga za tešnju Gisela 5), Malus (jabuka Gala Must) i Rubus (kupina Čačanska bestrna). AB  - Čuvanje biljnog materijala na izuzetno niskim temperaturama (tečni azot, -196°C) je veoma značajana metoda za dugotrajnu konzervaciju biljne germplazme. Pošto krioprezervacija značajno snižava troškove i rizike vezane za održavanje biljnih kolekcija u poljskim uslovima, ova metoda je prepoznata kao komplementarana konvencionalnim metodama čuvanja. Program primene ovakve strategije u očuvanju biljnih genetičkih resursa je iniciran i u Institutu za voćarstvo, Čačak. Istraživanja su fokusirana na optimizaciju protokola za različite tehnike krioprezervacije koje se baziraju na fenomenu vitrifikacije i na njihovu primenu u konzervaciji germpazme kontinentalnih voćaka. Tehnika inkapsulacije-dehidracije je uspešno primenjena u krioprezervaciji džanarike, kupine Čačanska bestrna i maline ‘Latham’. Ispitivanje primene tehnike vitrifikacija u konzervaciji germplazme Prunus-a i Malus-a je pokazalo da se značajno povećanje kapaciteta regeneracije krioprezerviranih vrhova izdanaka podloge za trešnju ‘Gisela 5’ i jabuke ‘Gala Must’ može postići variranjem dužine dehidracije i/ili vrste korišćenih vitrifikacionih rastvora. Optimizacija protokola za tehniku „droplet“ vitrifikacije kod predstavnika rodova Prunus (šljive Crvena ranka, Sitnica, Požegača, Krina, Džanarika i podloga ‘Gisela 5’), Malus (jabuka ‘Gala Must’) i Rubus (kupina Čačanska bestrna) je postignuta ispitivanjem uticaja različitih vitrifikacionih rastvora, dužine tretmana, temperature dehidracije i dužine rehidracije na ponovno rastenje krioprezerviranih eksplantata. Poslednjih godina, inicirana su takođe i istraživanja primene novo razvijenih „V cryo-plate“ i „D cryo-plate“ metoda krioprezervacije. Pored krioprezervacije in vitro eksplantata, u Institutu se vrše i ispitivanja uticaja kriotretmana na reproduktivnu sposobnost polena in vitro i in vivo kod različitih sorti šljive (Valjevka, Valerija, Čačanska lepotica) i trešnje (‘Kordia’, ‘Summit’).

Korišćenjem ovih metoda uspešno su krioprezervirani vrhovi in vitro izdanaka različitih autohtonih šljiva (Crvena ranka, Požegača, Sitnica, Moravka, Belošljiva), džanarike, podloge ‘Gisela 5’, jagode ‘Clery’, borovnice ‘Toro’ i mongolske borovnice. Pored krioprezervacije in vitro eksplantata, u Institutu se vrše i ispitivanja uticaja kriotretmana na reproduktivnu sposobnost polena in vitro i in vivo kod različitih sorti šljive (Valjevka, Valerija, Čačanska lepotica) i trešnje (‘Kordia’, ‘Summit’)., Storage of plant material at ultra-low temperature (liquid nitrogen, -196°C), has become important tool for long-term conservation of plant germplasm. Since cryopreservation reduces cost and risks linked to management of field germplasm collections, it has become а complementary approach to conventional conservation methods. Program for application of this strategy has also been initiated within Department of Fruit Physiology of FRI, Čačak. Most of the research is focused on optimization of the protocols for different vitrification-based cryopreservation techniques and their application for conserving fruit tree genotypes. Encapsulation-dehydration technique has been successfully applied for cryopreservation of myrobalan, blackberry ‘Čačanska Bestrna’ and raspberry ‘Latham’. Research on application of vitrification technique for conservation of Prunus and Malus germplasm has shown that significant increase in regrowth of cryopreserved shoot tips of cherry rootstock ‘Gisela 5’ and apple ‘Gala Must’ can be achieved by varying duration of dehydration and/or type of vitrification solution (VS) employed. Optimization of the droplet-vitrification protocol in representatives of Prunus (plums ‘Crvena Ranka’, ‘Sitnica’, ‘Požegača’ and ‘Krina’, myrobalan, rootstock ‘Gisela 5’), Malus (apple ‘Gala Must’) and Rubus (blackberry ‘Čačanska Bestrna’) genera was performed by evaluating the effect of different VSs, treatment durations, temperature of dehydration and duration of unloading on recovery of cryopreserved explants. Recently, the research on application of newly developed V and D cryo-plate techniques has been initiated, too. These methods allow overcoming disadvantages of other vitrification procedures such as mistiming dehydration, damage and loss of material and manipulative problems. Shoot tips of number of autochthonous plums (‘Crvena Ranka’,‘ Požegača’, ‘Sitnica’, ‘Moravka’, ‘Belošljiva’), myrobalan, rootstock ‘Gisela 5’, strawberry ‘Clery’, blueberry ‘Toro’ and saskatoon were successfully cryopreserved using these methods. Besides cryopreservation of in vitro explants, the effect of cryo-treatment on pollen reproductive ability in vitro and in vivo has also been examined in different plum (‘Valjevka’, ‘Valerija’, ‘Čačanska Lepotica’) and sweet cherry (‘Kordia’, ‘Summit’) cultivars.",

Za razvoj metode za efikasno in vitro razmnožavanje ovih genotipova posebna pažnja je posvećena optimizaciji faze aklimatizacije tokom 2017. i 2018. godine. Ožiljene i neožiljene biljke su posađene u supstrat koji se sastojao od treseta i zeolita u odnosu 2:1, gde se pratilauspešnost aklimatizacije pod „mist” sistemom u staklari. U 2017. godini najoptimalniji mesec za aklimatizaciju ožiljenih i neožiljenih biljaka za obe sorte maline bio je jun , kada je uspešnost aklimatizacije ožiljenih biljaka bila 100% za obe sorte. U istom periodu, neožiljene biljke su aklimatizovane sa uspešnošću od 94,74% (‘Meeker‘) odnosno 97,56% (‘Willamette‘). U 2018. godini avgust je bio najoptimalniji mesec za aklimatizaciju ožiljenih biljaka kod sorte maline ‘Willamette‘ (99,55%), dok je kod sorte ‘Meeker‘ jul (100%). Kod obe sorte maline jul je bio najoptimalniji mesec za aklimatizaciju neožiljenih biljaka (96,54% kod ‘Willamette‘ i 98,33% kod ‘Meeker‘). Uzimajući u obzir ukupan broj ožiljenih i neožiljenih biljaka tokom dve eksperimentalne godine, uspešnost aklimatizacije ožiljenih biljaka kod sorte ‘Meeker‘ je 96,00% i 93,10% neožiljenih biljaka. Kod sorte maline ‘Willamette‘ uspešno je aklimatizovano 97,69% ožiljenih i 85,79% neožiljenih biljaka.
AB  - Micropropagation is a modern biotechnological method for the production of genetically identical, physiologically uniform and healthy plants without pathogens in the shortest possible time. Considering the problems in raspberry production in Serbia, Fruit Research Institute, Čačak initiated controlled production of ‘Meeker‘ and ‘Willamette‘ planting material by micropropagation using nuclear stock produced according to the Certification scheme for Rubus. To develop а method for efficient in vitro propagation of these genotypes special attention has been devoted to optimization of acclimatization stages during 2017 and 2018. Rooted and unrooted plants are placed in a substrate consisting of peat and zeolite in a ratio of 2:1 where the success of acclimatization under the "mist" system in the glassworks was monitored. In 2017, the most optimal month for acclimatization of rooted and unrooted shoots for both raspberries cultivars were June, when the success of acclimatization of rooted shoots was 100% for both cultivars. In the same period, unrooted shoots were acclimatized with a success rate of 94.74% (‘Meeker‘) and 97.56% (‘Willamette‘), respectively. In 2018, August was the most optimal month for acclimatization of rooted shoots in the raspberry cultivar ‘Willamette‘ (99.55%), while in the ‘Meeker‘ cultivar it was July (100%). For both raspberry cultivars, July was the most optimal month for acclimatization of unrooted shoots (96.54% for ‘Willamette‘ and 98.33% for ‘Meeker‘). Taking into account the total number of rooted and unrooted plants during the two experimental years, the success of acclimatization of rooted shoots in the ‘Meeker‘ cultivars 96.00% and 93.10% of unrooted shots. In the raspberry cultivar ‘Willamette‘, 97.69% of rooted and 85.79% of unrooted shoots were successfully acclimatized.

Optimization of the protocol for in vitro propagation of autochthonous plum genotype ‘Metlaš’

AB  - Fast and cost-effective clonal propagation of planting material is possible to achieve by application of tissue culture in vitro. In this way, problems associated with traditional propagation, such as rapid spreading of diseases, lack of initial material caused by dependence on seasonal growth or low propagation coefficient can be overcome. This paper deals with optimization of micropropagation of autochthonous plum ‘Metlaš’ (Prunus domestica L.) originated from Guberevci (Municipality Knić, Serbia). ‘Metlaš’ is very often considered to be the same genotype as ‘Okruglica’ (syn. ‘Dragačica’), but they can be clearly distinguished according to plant habit, stone and fruit characteristics. To optimize multiplication stage, the influence of benzyladenine (BA at 0.5, 1.0, 1.5 and 2.0 mg/l) and thidiazuron (TDZ at 0.25, 0.5, 1.0 and 1.5 mg/l) on the multiplication capacity (multiplication index, length of axial and lateral shoots) was examined. Rooting ability of shoots (rooting rate, number and length of roots, and height of rooted plants) was monitored on half strength Murashige and Skoog (MS) medium containing 1.0 mg/l indole-3-butyric acid (IBA) or 1-naphthaleneacetic acid (NAA), each combined with 0.1 mg/l gibberellic acid (GA3). TDZ applied at 0.25 mg/l gave the highest multiplication index (9.1), while the longest axial (14.1 mm) and axillary shoots (8.6 mm) were obtained on medium with 1.5 mg/l BA. Although both auxins proved to be efficient in rhizogenesis (rooting rates being 85.7% and 95.2%), higher values of all rooting parameters were observed in the presence of IBA. Rooted shoots were successfully acclimatized.

AB  - Krioprezervacija polena omogućava ukrštanja između genotipova koji se razlikuju po vremenu cvetanja, odnosno gajenih na udaljenim lokacijama, što je od velikog značaja za korišćenje germplazme u oplemenjivačke svrhe, kao i očuvanje genetičkih resursa voćaka. Osnovni cilj istraživanja je bio utvrđivanje uticaja kriotretmana (temperatura tečnog azota) na reproduktivnu sposobnost polena monitoringom rasta polenovih cevčica in vivo u tučkovima trešnje (Prunus avium L.). Uticaj kriotretmana je razmatran u kontekstu njegove interakcije sa genotipom oprašivača, i mogućnosti koje pruža analiza in vivo parametara rasta polenovih cevčica pri proceni reproduktivne sposobnosti polena nakon krioprezervacije. Tučkovi sorte trešnje ‘Reginaʼ oprašeni su kriotretiranim i netretiranim polenom sorti ‘Kordiaʼ i ‘Summitʼ. Metodom fluorescentne mikroskopije određeni su kvantitativni parametri rasta in vivo (brojnost polenovih cevčica u pojedinim regionima tučka, dinamika rasta polenovih cevčica, procenat oplođenja), a dobijeni podaci su analizirani zajedno sa podacima utvrđenim za in vitro parametre rasta, kao i zametanje plodova. Rezultati istraživanja upućuju na sledeće zaključke: i) reproduktivna sposobnost polena se može uspešno testirati analizom in vivo parametara rasta polenovih cevčica, koju treba kombinovati sa in vitro testom; ii) pri proceni kvaliteta kriotretiranog polena treba koristiti semi- ili potpuno kompatibilne majčinske sorte, čiji sporofiti podržavaju rast muškog gametofita; iii) ključni parametri in vivo testa za procenu reproduktivne sposobnosti kriotretiranog polena su broj polenovih cevčica u gornjoj trećini stubića i plodniku, kao i procenat oplođenja (procenat tučkova sa prodorom najduže polenove cevčice u nucelus semenog zametka desetog dana od dana oprašivanja). Reproduktivna sposobnost polena trešnje nakon krioprezervacije je očuvana u značajnoj meri, i može se proceniti primenom in vivo testa sa značajnom verodostojnošću, u roku od najviše dve nedelje nakon odmrzavanja i rehidracije.
AB  - Cryopreservation of fruit species pollen allows cross-breeding between genotypes that differ in flowering time, or between the plants grown in distant places, affecting the use of germplasm for breeding purposes and conservation of fruit genetic resources. The primary objective of our research was to determine effect of cryotreatment on pollen reproductive ability in vivo by monitoring pollen tubes growth in the sweet cherry (Prunus avium L.) pistils. The influence of cryotreatment was considered from the context of its interaction to polleniser genotype, and possibility for usage of pollen performance in vivo parameters for assessment of pollen fertilization ability after cryopreservation. Pistils of sweet cherry cultivar ‘Regina’ were pollinated with both cryotreated and non-cryotreated pollen of ‘Kordia’ and ‘Summit’. Quantitative parameters of pollen tube growth in vivo (pollen tube number in the certain pistil parts, pollen tube dynamics, fertilization percentage) were determined using fluorescence microscopy method, and obtained data were analyzed together with those obtained for in vitro pollen performance and fruit set. The research have indicated that: i) reproductive ability of cryotreated sweet cherry pollen is verifiable analyzing in vivo parameters, which should be combined with in vitro test; ii) semi- or fully-compatible female recipients, whose sporophytes are supportive for male gametophytes should be used for the assessment; iii) the key parameters for in vivo testing are pollen tube number in the upper part of the style and in the ovary, as well as fertilization percentage, i.e. the percentage of pistils with penetration of the longest pollen tube in the nucellus of the ovary on the 10th day after pollination. The assessment of pollen reproductive ability after cryopreservation using in vivo testing can give the answers about pollen quality with valuable credibility, in maximum two weeks after thawing and rehydration.

Osnovni cilj istraživanja je bio utvrđivanje uticaja kriotretmana (temperatura tečnog azota) na reproduktivnu sposobnost polena monitoringom rasta polenovih cevčica in vivo u tučkovima trešnje (Prunus avium L.). Uticaj kriotretmana je razmatran u kontekstu njegove interakcije sa genotipom oprašivača, i mogućnosti koje pruža analiza in vivo parametara rasta polenovih cevčica pri proceni reproduktivne sposobnosti polena nakon krioprezervacije. Tučkovi sorte trešnje ‘Reginaʼ oprašeni su kriotretiranim i netretiranim polenom sorti ‘Kordiaʼ i ‘Summitʼ. Metodom fluorescentne mikroskopije određeni su kvantitativni parametri rasta in vivo (brojnost polenovih cevčica u pojedinim regionima tučka, dinamika rasta polenovih cevčica, procenat oplođenja), a dobijeni podaci su analizirani zajedno sa podacima utvrđenim za in vitro parametre rasta, kao i zametanje plodova. Rezultati istraživanja upućuju na sledeće zaključke: i) reproduktivna sposobnost polena se može uspešno testirati analizom in vivo parametara rasta polenovih cevčica, koju treba kombinovati sa in vitro testom; ii) pri proceni kvaliteta kriotretiranog polena treba koristiti semi- ili potpuno kompatibilne majčinske sorte, čiji sporofiti podržavaju rast muškog gametofita; iii) ključni parametri in vivo testa za procenu reproduktivne sposobnosti kriotretiranog polena su broj polenovih cevčica u gornjoj trećini stubića i plodniku, kao i procenat oplođenja (procenat tučkova sa prodorom najduže polenove cevčice u nucelus semenog zametka desetog dana od dana oprašivanja). Reproduktivna sposobnost polena trešnje nakon krioprezervacije je očuvana u značajnoj meri, i može se proceniti primenom in vivo testa sa značajnom verodostojnošću, u roku od najviše dve nedelje nakon odmrzavanja i rehidracije., Cryopreservation of fruit species pollen allows cross-breeding between genotypes that differ in flowering time, or between the plants grown in distant places, affecting the use of germplasm for breeding purposes and conservation of fruit genetic resources. The primary objective of our research was to determine effect of cryotreatment on pollen reproductive ability in vivo by monitoring pollen tubes growth in the sweet cherry (Prunus avium L.) pistils. The influence of cryotreatment was considered from the context of its interaction to polleniser genotype, and possibility for usage of pollen performance in vivo parameters for assessment of pollen fertilization ability after cryopreservation. Pistils of sweet cherry cultivar ‘Regina’ were pollinated with both cryotreated and non-cryotreated pollen of ‘Kordia’ and ‘Summit’. Quantitative parameters of pollen tube growth in vivo (pollen tube number in the certain pistil parts, pollen tube dynamics, fertilization percentage) were determined using fluorescence microscopy method, and obtained data were analyzed together with those obtained for in vitro pollen performance and fruit set. AB  - A study was conducted to investigate the effect of different storage periods and temperatures on pollen viability in vitro and in vivo in plum genotypes 'Valerija', 'Cacanska Lepotica' and 'Valjevka'. In vitro pollen viability was tested at day 0 (fresh dry pollen) and after 3, 6, 9 and 12 months of storage at four different temperatures (4, -20, -80 and -196 degrees C), and in vivo after 12 months of storage at distinct temperatures. In vitro germination and fluorescein diacetate (FDA) staining methods were used to test pollen viability, while aniline blue staining was used for observing in vivo pollen tube growth. Fresh pollen germination and viability ranged from 42.35 to 63.79% ('Valjevka' and 'Cacanska Lepotica', respectively) and 54.58 to 62.15%, ('Valjevka' and 'Valerija', respectively). With storage at 4 degrees C, pollen viability and germination decreased over the period, with the lowest value after 12 months of storage. Pollen germination and viability for the other storage temperatures (-20, -80 and -196 degrees C) were higher than 30% by the end of the 12 months. Pollination using pollen stored at 4 degrees C showed that pollen tube growth mostly ended in the lower part of the style. With the other storage temperatures, pollen tube growth was similar, ranging between 50 and 100% of the pistils with pollen tubes penetrated into the nucellus of the ovule in the genotype 'Cacanska Lepotica'. The results of these findings will have implications for plum pollen breeding and conservation.

AB  - Protokole krioprezervacije potrebno je testirati za autohtone sorte na zaraženom i zdravom sadnom materijalu, jer zaraženost može predstavljati problem u kulturi tkiva. Antioksidansi mogu potaknuti rast biljaka. Cilj rada bio je uspostaviti regeneraciju biljaka inficiranih genotipova u in vitro uvjetima, te na njima testirati standardni protokol krioprezervacije za vinovu lozu. Pupovi inficiranih genotipova veličine 1,5 mm postavljeni su na različite MS (Murashige i Skoog) medije uz dodatak 0,5 mg/L 6-benzilaminopurina (BAP) i 0,05 mg/L indol-3-octene kiseline (IAA) sa antioksidansima (M1: 0,1 mM salicilne kiseline, M2: 1 mM askorbinske kiseline, M3: 1 mM glutationa i M4:kombinacija tri antioksidansa navedenih koncentracija), te je praćen njihov rast in vitro. Iz potjeralih biljaka izolirani su vršni meristemi, te je proveden pokus krioprezervacije (LS-20´, ½ PVS2-30´i PVS2-50´ kontrola i smrzavanje). Najveći rast in vitro postignut je na mediju sa dodatkom glutationa (M3). Zaraženost je imala utjecaj u regeneraciji biljaka in vitro, ali nije utjecala na regeneraciju nakon krioprezervacije.
AB  - Cryopreservation protocols need to be tested for autochthonous cultivars on an infected and healthy plant material cause virus infection can be difficult in tissue culture. Antioxidants can improve the plant growth. The aim of this work was to establish the plant regeneration of infected plants in tissue culture conditions and testing a standard cryopreservation protocol for grapevine on them. Buds 1.5.mm-size of infected genotypes were inoculated on different MS (Murashige i Skoog) media with addition of 0.5 mg/L 6-benzilaminopurina (BAP) and 0.05 mg/L indol-3-acetic acid (IAA) with antioxidants (M1: 0.1 mM salycilic acid, M2: 1 mM ascorbic acid, M3: 1 mM glutathione and M4: combination of three antioxidants with mentioned concentrations) and in vitro growth was observed. From shooted plants apical meristems were excised and cryopreservation protocol was performed (LS-20´, ½ PVS2-30´and PVS2-50´ control and freezing). The highest in vitro growth was achieved on medium with addition of glutathione (M3). Infection has an effect on regeneration of plants, but has not effect on regeneration after cryopreservation.

Morphological and pathogenic characteristics of Alternaria alternata isolates from plum (Prunus domestica L.)

AB  - During June and July 2020, numerous ‘Stanley’ plants were observed with small, brown spots on leaves in the plum orchard in locality Bačka Topola. Later on, leaf lesions became dark brown and irregular, resulting in necrosis that was followed by defoliation. Collected samples were analyzed using standard mycological procedures. Ten fungal isolates from leaves and branches were obtained from infected plums. Two isolates originating from branches (ŠG) and leaves (ŠL), were selected for further studies. The pathogenicity test was performed on five plum
cultivars: ‘Stenley’, ‘Čačanska Lepotica’, ‘Čačanska Rodna’, ‘Čačanska Najbolja’ and ‘Požegača’. Morphological characteristics of two selected isolates were studied on three nutrient media: cornmeal agar (CMA), carnation leaf agar (CLA) and potato dextrose agar (PDA). Both tested isolates formed airy, dark olive-green colonies on PDA medium. The colonies were light olive-green in all tested isolates on CMA. On CLA, all studied isolates formed olive-green substrate mycelium. Conidia size for tested isolate ŠG on PDA medium ranged from 19.20−46.24 × 7.95−12.88 μm (mean 25.85 × 10.42 μm), and for isolate ŠL from 18.17−36.16 × 8.09−12.59 μm, (mean 24.98 × 11.10 μm). Both studied isolates on all tested plant organs (leaf, branch and fruit) in all plum cultivars caused symptoms typical for fungi of the genus Alternaria, the appearance of dark brown necrotic lesions on leaves, branches and fruits. Based on preliminary research of morphological characteristics and pathogenic tests, analyzed isolates were identified as belonging to the species Alternaria alternata.

Ten fungal isolates from leaves and branches were obtained from infected plums. Two isolates originating from branches (ŠG) and leaves (ŠL), were selected for further studies. The pathogenicity test was performed on five plumcultivars: ‘Stenley’, ‘Čačanska Lepotica’, ‘Čačanska Rodna’, ‘Čačanska Najbolja’ and ‘Požegača’. Morphological characteristics of two selected isolates were studied on three nutrient media: cornmeal agar (CMA), carnation leaf agar (CLA) and potato dextrose agar (PDA). Both tested isolates formed airy, dark olive-green colonies on PDA medium. The colonies were light olive-green in all tested isolates on CMA. On CLA, all studied isolates formed olive-green substrate mycelium. Conidia size for tested isolate ŠG on PDA medium ranged from 19.20−46.24 × 7.95−12.88 μm (mean 25.85 × 10.42 μm), and for isolate ŠL from 18.17−36.16 × 8.09−12.59 μm, (mean 24.98 × 11.10 μm). Both studied isolates on all tested plant organs (leaf, branch and fruit) in all plum cultivars caused symptoms typical for fungi of the genus Alternaria, the appearance of dark brown necrotic lesions on leaves, branches and fruits. Based on preliminary research of morphological characteristics and pathogenic tests, analyzed isolates were identified as belonging to the species Alternaria alternata.",

Of all analyzed pathogens, only PPV was detected in six samples. The presented results showed that, even in the high virus inoculum pressure, it is possible to select virus-free trees from the existing old orchards. After analysis, selected healthy trees were used as a source of explants for the establishment of aseptic culture of each genotype for the conservation purpose.

AB  - The aim of this study was to evaluate the content and profile of the phenolic compounds (PCs) and antioxidant properties of field-grown leaves, in vitro leaves and in vitro callus cultures of the blackberry 'Cacanska Bestrna' and blueberry 'Toro'. In vitro shoots of the selected genotypes were grown either on original Murashige and Skoog (MS) medium containing 1 mg/L BA, 0.1 mg/L IBA and 0.1 mg/L GA(3) ('Cacanska Bestrna') or on MS medium with macroelements reduced to 1/2, 2 mg/L zeatin and 0.2 mg/L IAA ('Toro'). Callus cultures were induced from in vitro leaves and established on MS medium with 2 mg/L BA and 2 mg/L 2,4-D ('Cacanska Bestrna') or MS medium with half strength macroelements, 2 mg/L BA, 2 mg/L 2,4-D and 1 mg/L NAA ('Toro'). Total phenolic (TPC) and flavonoid content (TFC) were the highest in blueberry leaves, whereas low TPC and TFC values were obtained in callus cultures of both cultivars. A higher content of PCs in blueberry leaves compared to blackberry leaves was determined by the UHPLC-DAD MS/MS technique. Quercetin derivatives and phenolic acids were the dominant PCs in the leaves of both berries, whereas gallocatechin was present in a significant amount in blueberry leaves. Callus cultures of both berries had a specific PC profile, with none detected in the leaves except quercetin-3-O-glucoside and quercetin-3-O-rutinoside. Blackberry leaves showed the best antioxidant properties as estimated by ferric reducing power (FRP), ABTS(& BULL;+) and DPPH & BULL; scavenging activity assays. Callus cultures of both berries exhibited three to five times lower ABTS(& BULL;+) and ten to seventeen times lower DPPH & BULL; scavenging activity compared to corresponding leaves. The analyzed leaves and callus cultures can be a good source of PCs with good antioxidant properties and specific phenolics, respectively, for applications in the food and pharmaceutical industries.

Reproductive ability of plum (Prunus domestica L.) pollen stored at low temperatures

AB  - Breeding programmes often require the necessity for longer pollen storage, whether asynchronous flowering of parental pairs exists (especially if early-flowering genotypes are pollinated with late-flowering), or if there is a temporal, geographical or spatial distance. The objective of this study was to assessed pollen vitality in vitro and in vivo in three plum cultivars (‘Čačanka Lepotica’, ‘Valjevka’ and ‘Valerija’) after one year of storage at +4°C and sub-freezing temperatures (-20°C, -80°C and -196°C). Pollen viability in vitro was determined on germination medium and by staining with fluorescein-diacetate. For in vivo vitality assay, stored pollen was used for pollination of emasculated flowers of ‘Čačanka Lepotica’. Parameters of pollen tubes growth was determined on 3th, 6th and 10th day after pollination. In all cultivars, the lowest rate of pollen viability was observed for that stored at +4°C. Pollen stored at sub-freezing temperatures showed relatively high viability that was reduced from 8.85 to 47.15% in comparison with fresh pollen. Generaly, in vitro germination test showed lower pollen viability compared to fluorescein-diacetate staining method. Pollen viability under in vivo conditions, corellated with the those obtained under in vitro conditions. The growth of pollen tubes, from pollen stored at +4°C, or was absent or it ended mainly in the upper parts of the style. For pollen stored at sub-freezing temperatures, pollen tubes penetrated into the nucelus on the 6th day after pollination in all cultivars. The best dynamic of pollen tube growth of ‘Čačanska Lepotica’ and ‘Valerija’ was observed with pollen stored at -80°C, while in ‘Valјevka’ with that stored at -196°C.

abstract = "Breeding programmes often require the necessity for longer pollen storage, whether asynchronous flowering of parental pairs exists (especially if early-flowering genotypes are pollinated with late-flowering), or if there is a temporal, geographical or spatial distance. The objective of this study was to assessed pollen vitality in vitro and in vivo in three plum cultivars (‘Čačanka Lepotica’, ‘Valjevka’ and ‘Valerija’) after one year of storage at +4°C and sub-freezing temperatures (-20°C, -80°C and -196°C). Pollen viability in vitro was determined on germination medium and by staining with fluorescein-diacetate. For in vivo vitality assay, stored pollen was used for pollination of emasculated flowers of ‘Čačanka Lepotica’. Parameters of pollen tubes growth was determined on 3th, 6th and 10th day after pollination. In all cultivars, the lowest rate of pollen viability was observed for that stored at +4°C. Pollen stored at sub-freezing temperatures showed relatively high viability that was reduced from 8.85 to 47.15% in comparison with fresh pollen. Generaly, in vitro germination test showed lower pollen viability compared to fluorescein-diacetate staining method. Pollen viability under in vivo conditions, corellated with the those obtained under in vitro conditions. The growth of pollen tubes, from pollen stored at +4°C, or was absent or it ended mainly in the upper parts of the style. For pollen stored at sub-freezing temperatures, pollen tubes penetrated into the nucelus on the 6th day after pollination in all cultivars. The best dynamic of pollen tube growth of ‘Čačanska Lepotica’ and ‘Valerija’ was observed with pollen stored at -80°C, while in ‘Valјevka’ with that stored at -196°C.",

Cryopreservation of Serbian autochthonous plum 'Crvena ranka' using aluminium cryo-plates

'Crvena Ranka' is one of the oldest autochthonous plum cultivars in Serbia. However, it is endangered due to the long-term pathogen pressure and continuous suppression by commercially important cultivars and therefore faced with genetic erosion. This study was carried out to investigate the suitability of two efficient and simple cryopreservation methods using aluminium cryo-plates for its conservation. Precultured shoot tips of this genotype were embedded in alginate gel into cryo-plates wells. Osmoprotection was performed using loading solution with 1.9 M glycerol and 0.5 M sucrose (30 min at room temperature). In the V cryo-plate protocol, explants were dehydrated at room temperature for 20 or 40 min with PVS A3 (37.5% glycerol, 15% dimethylsulfoxide, 15% ethylene glycol and 22.5% sucrose) or with PVS3 (50% glycerol and 50% sucrose) for 60 min. In the D cryo-plate protocol, desiccation for 2, 2.5 or 3 h was performed over silica gel. Then, the cryo-plates were directly immersed into liquid nitrogen. Unloading was done in MS medium containing 0.8 M sucrose (30 min at room temperature). In the V cryo-plate procedure regrowth of cryopreserved explants dehydrated with PVS A3 was between 50% and 51.9%, while in those dehydrated with PVS3 it was 66.7%. As for the D cryo-plate method, regrowth of cryopreserved explants ranged between 30-40%. After regrowth, shoots were successfully multiplied and rooted. Results prove the feasibility of these new cryogenic methods for a long-term storage of this valuable Prunus genotype.

Morphological and pathogenic characteristics of Alternaria alternata isolates from plum (Prunus domestica L.)

AB  - During June and July 2020, numerous 'Stanley' plants were observed with small, brown spots on leaves in the plum orchard in locality Bačka Topola. Later on, leaf lesions became dark brown and irregular, resulting in necrosis that was followed by defoliation. Collected samples were analyzed using standard mycological procedures. Ten fungal isolates from leaves and branches were obtained from infected plums. Two isolates originating from branches (ŠG) and leaves (ŠL), were selected for further studies. The pathogenicity test was performed on five plum cultivars: 'Stanley', 'Čačanska Lepotica', 'Čačanska Rodna', 'Čačanska Najbolja' and 'Požegača'. Morphological characteristics of two selected isolates were studied on three nutrient media: cornmeal agar (CMA), carnation leaf agar (CLA) and potato dextrose agar (PDA). Both tested isolates formed airy, dark olive-green colonies on PDA medium. The colonies were light olive-green in all tested isolates on CMA. On CLA, all studied isolates formed olive-green substrate mycelium. Conidia size for tested isolate ŠG on PDA medium ranged from 19.20 to 46.24×7.95-12.88 μm (mean 25.85×10.42 μm), and for isolate ŠL from 18.17 to 36.16×8.09-12.59 μm, (mean 24.98×11.10 μm). Both studied isolates on all tested plant organs (leaf, branch and fruit) in all plum cultivars caused symptoms typical for fungi of the genus Alternaria, the appearance of dark brown necrotic lesions on leaves, branches and fruits. Based on preliminary research of morphological characteristics and pathogenic tests, analyzed isolates were identified as belonging to the species Alternaria alternata.
