AB  - Nova sorta višnje ‘Sofija’, priznata je rešenjem Ministarstva polјoprivrede i zaštite životne sredine RS br. 320-04-00566/2011-11 od 19. januara 2015. godine. Stvorena planskom hibridizacijom iz ukrštanja ‘Čačanski rubin’ × ‘Heimanns Konserven Weichsel’, ova sorta je izdvojena u grupu od deset elitnih hibrida pod oznakom XII/57, iz populacije od preko 3.000 hibridnih sejanaca. Stablo sorte ‘Sofija’ je umerene bujnosti, sa karakterističnim povijenim granama, srednje ranog cvetanja. Plodovi su veoma krupni (7‒8 g), zaoblјenog oblika, slabo izraženog šava. Pokožica ploda je rubin crvene boje, glatka i sjajna, a mezokarp crvene boje, slatko-nakiselog ukusa i odličnog kvaliteta. Sok je crvene boje. Koštica je srednje krupnoće, a petelјka izrazito duga, tanka i lako se odvaja od ploda, bez curenja soka. ‘Sofija’ se odlikuje visokim stepenom polјske otpornosti na Blumeriella jaapii (Rehm) v. Arx. Ono što je od posebnog značaja za ovu sortu jeste njeno veoma interesantno vreme sazrevanja (početak druge dekade juna u uslovima zapadne Srbije). Imajući u vidu činjenicu da aktuelni sortiment višnje nije previše dinamičan, kao i da je vreme sazrevanja najvećeg broja komercijalno značajnih krupnoplodih sorti vezano za treću dekadu juna, plodovi sorte ‘Sofija’ zbog ranijeg vremena sazrevanja, uz istovremeno izvanredan kvalitet ploda i visoku rodnost, ostvaruju visoku cenu na tržištu. Od 2019. godine, sorta ‘Sofija’ je standardni deo asortimana u proizvodnji sadnog materijala višnje u Republici Srbiji, a od 2023. godine i van granica naše zemlje.
AB  - The new sour cherry cultivar ‘Sofija’ was named and released by the Ministry of Agriculture, Forestry, and Water Management of the Republic of Serbia (confirmation No. 320-04-00566/2011-11; January 11th, 2015). Developed by planned hybridization from the crossing of ‘Čačanski rubin’ × ‘Heimanns Konserven Weichsel’, this cultivar was selected into the group of ten elite hybrids (under the designation XII/57), from a population of over 3,000 hybrids. The tree of the ‘Sofia’ cultivar is moderately vigorous, with characteristic bent branches, of medium-early flowering. The fruits are very large (7‒8 g), rounded, with a weak seam. The skin of the fruit is ruby red in color, smooth and shiny, and the mesocarp is red, with a sweet-sour taste and excellent quality. The juice is red. The stone is of medium size, and the stem is very long, thin and easily separated from the fruit, without leaking juice. ‘Sofia’ is characterized by a high degree of field resistance to Blumeriella jaapii (Rehm) v. Arch. What is of special importance for this cultivar is its very interesting ripening time (the beginning of the second decade of June in the conditions of western Serbia). Bearing in mind the fact that the current sour cherry assortment is not too dynamic, as well as that the ripening time of the largest number of commercially significant large-fruited cultivars is related to the third decade of June, the fruits of the ‘Sofia’ cultivar, due to their earlier ripening time, with at the same time outstanding fruit quality and high fertility, achieve high price on the market. Since 2019, the variety ‘Sofija’ is a standard part of the assortment in the production of cherry planting material in the Republic of Serbia, and from 2023 also outside the borders of our country.

AB  - In this study performance in vitro of fresh and pollen stored for three months at different constant temperatures were investigated in three plum cultivars (ʻValerijaʼ, ʻČačanska Lepoticaʼ, ʻValjevkaʼ). Pollen were collected from the flowers in late balloon phase and after drying stored for three months at four different constant temperatures (+4 °C, ‒20 °C, ‒80 °C, ‒196 °C). Pollen tube length was examined as well as the nuclear presence in pollen tube and its migration. In vitro pollen tube length was tested after a 24-hour germination period after which the pollen tube was stained with Hoechst 33258 to observe the nuclear presence within it. The longest average pollen tube length in all tested cultivars were observed with fresh pollen. With the pollen stored at low temperatures the longer pollen tube was obtain compared to pollen stored at +4 °C. In all cultivars mature pollen grains were in binucleate stage contained vegetative nucleus (VN) and generative nucleus (GN). Although at each storage temperature a higher percentage of GN was achieved in pollen tubes compared to fresh ones, nevertheless, regular movement of nuclei, primarily VN than GN or sperm nucleus (SN), was found in the highest percentage in all tested cultivars.
AB  - U ovom radu su ispitane performanse in vitro svežeg i polena čuvanog na različitim konstantnim temperaturana tokom tri meseca kod tri sorte šljive (ʻValerijaʼ, ʻČačanska Lepoticaʼ, ʻValjevkaʼ). Polen je sakupljan iz cvetova koji su bili u kasnoj balon fazi i nakon sušenja čuvan na četiri konstantne temperature (+4 °C, ‒20 °C, ‒80 °C, ‒196 °C). Ispitana je dužina polenovih cevčica, prisustvo i migracija jedara u njima. Dužina polenovih cevčica je određivana nakon inkubacionog perioda od 24h a zatim bojenje sa Hoechst 33258 da bi se uočilo prisustvo jedara u njima. Prosečno najduže polenove cevčice kod svih ispitivanih sorti utvrđene su kod svežeg polena. Kod polena čuvanog na niskim temperaturama prosečna dužina polenovih cevčica je bila veća nego kod polena čuvanog na +4 °C. Polen svih sorti u zrelom stanju je dvojedaran, sadrži vegetativno i generativno jedro. Iako je kod svih sorti nakon perioda čuvanja polena, uočen veći procenat prisustva genetativnog jedra u polenovim cevčicama, raspored jedara je bio regularan, prvo vegetativno, potom generativno ili spermatično jedro.

Breeding work and floral biology research in cherries at Fruit Research Institute, Cacak – achieved results and new perspectives

AB  - Decades-long cherry breeding work at Fruit Research Institute, Čačak (FRI), implies the use as a main method planned hybridization within P. avium and P. cerasus. Combined with the selection from the natural populations, the method resulted in realization of two sweet cherry ('Asenova Rana' and 'Čarna') and five sour cherry cultivars ('Čačanski Rubin', 'Šumadinka', 'Sofija', 'Nevena' and 'Iskra'), three hybrids in the procedure of recognition, and numerous being intensively studied. Current sour cherry breeding work is based on the use of domestic genotypes, well adapted to the environmental conditions of the area (either obtained by planned hybridization or indigenous), and foreign genotypes – tolerant to diseases, with high cropping potential and good fruit quality. Different aspects of reproductive biology in cherries – flowering phenology, pollen tube growth in vitro and in vivo (pollen tube growth dynamics, pollen-pistil interactions in the style and ovary), cytoembryology (ovule development stage, viability of ovule and embryo sac, early embryogenesis), are among the main research activities at FRI. Within the past decade, the research on gametofitic self-incompatibility (GSI) in cherries has been advanced by identification of S-haplotypes in cultivars, landraces and hybrids using consensus, specific and dCAPS primers to reveal S-RNase and SFB functional/non-functional variants. Besides the GSI, other possible types of pollen-pistil interactions have also been considered, primarily in the context of temperature conditions during the flowering. Nowadays, within the CherrySeRB project whose FRI is the leading institution, characterization of the indigenous genotype potential for desirable reproductive properties and defining ʻgood reproductive behaviour cherry modelʼ that can face warmer temperature conditions during the flowering are in focus. The main CherrySeRB idea is connection of production challenges arising from climate change and the richness of Serbian and Balkan cherry germplasm that is still unused enough, which represents a novelty in breeding approach, applicable to other fruit species.

AB  - In sour cherry (Prunus cerasus L.), as in other species of the Prunus genus, the phenomenon of gametophytic self-incompatibility occurs. Basically similar to another Rosaceae species, sour cherry GSI is quite complex due to its tetraploid nature and the presence of pollen-part and stylar-part mutants that cause genetic changes at the S-locus. The phenomenon implies self-incompatible, partially self-compatible and self-compatible reproductive behavior of cultivars, as well as requirement for pollenisers in commercial orchards. Self-(in)compatibility status of sour cherry cultivars/selections realized at Fruit Research Institute, Čačak [‘Šumadinka’ (‘Köröser Weichsel’ × ‘Heimanns Konserven Weichsel’), ‘Sofija’ (‘Čačanski Rubin’ × ‘Heimanns Konserven Weichsel’), ‘Nevena’ (‘Köröser Weichsel’ × ‘Heimanns Konserven Weichsel’) and ‘GV-6’ (selected from natural population in the Čačak vicinity)] was examined over two years, and compared to standard ‘Heimanns Konserven Weichsel’. Self-pollination experiment in the field was conducted, as well as monitoring the parameters of pollen tubes growth efficacy in vivo by fluorescent microscopy (pollen tubes number and kinetics, fertilization percentage and presence of incompatible pollen tubes), and fruit set recording. In the pistils of ‘Nevena’, pollen tubes ended their growth predominantly in the upper part of the style, with sporadically penetrated tubes in the middle style. Pollen tubes kinetics was more efficient in the styles of other genotypes ‒ they were in the micropyle already on the 3rd day, and in the nucellus of the ovary on the 6th day after pollination (‘Šumadinka’, ‘Heimanns Konserven Weichsel’ and ‘Sofija’). According to fertilization percentage and fruit set, ‘Nevena’ could be undoubtedly classified within self-incompatible cultivars (both parameters were 0.00%, in both seasons), whereas ‘Šumadinka’ (17.92%; 16.31%, respectively) and ‘Heimanns Konserven Weichsel’ (18.75%; 17.50%, respectively) behaved as self-compatible. Showing relatively high values of fertilization percentage (21.05%; 23.18%, respectively), but lower values of fruit set (11.57%; 7.24%) after selfing, ‘Sofija’ and ‘GV-6’ could be classified as partially self-compatible genotypes.

Influence of temperature at the time of pollination on the effective pollination period and fruit set in plum

AB  - So far, the concept of effective pollination period (EPP) for several cultivars of plum, apricot, apple, pear, almond and others has been proven as suitable parameter to set up a relationship between the variation in the reproductive process and cropping behaviour. Among a number of autochthonous plum cultivar present in plum orchards in Serbia, ‘Crvena Ranka’ is the most important. It’s a self-sterile cultivar (characterized by the appearance of cytoplasmic male sterility), so the presence of an adequate pollenizer is needed. Fruit set and EPP in this plum cultivar were evaluated under natural field conditions in western Serbia during 2021 and 2022. Pollination of flowers with mix of dry pollen was subsequenly done during the first 10 days after full bloom. Initial and final fruit set was recorded four weeks after pollination and at the period of physiological maturity of the fruit, respectively. Results show that in both years number of initial fruit set increased up to five days after anthesis. The same was archived with the final fruit set in the first year while in the second, the higest number of fruit set was observed on the fourth day after anthesis (40.67% and 54.48%, resp.). In the first year, five days after anthesis a slightly deacrease in final fruit set was observed with the lowest, but still satisfactory values, on the 10th day after anthesis (11%). In the second year, decreasing in fruit set was more evident and was below 4% on the seventh day after anthesis. The reason for such significant decrease in fruit set was the spring frost occurred during the five days in a row. Taking into account clearly effects of weather conditions on fruit set, based on obtained results, it can be estimated that the EPP for plum cultivar ‘Crvena Ranka’  is 10 days.

AB  - Značajan napredak u oplemenjivanju koštičavih vrsta voćaka ostvaren je širom sveta, u cilju dobijanja novih sorti prilagođenih različitim agro-ekološkim uslovima, sa karakteristikama koje obezbeđuju komercijalne efekte i odgovaraju zahtevima potrošača u pogledu kvaliteta i zdravstvene bezbednosti plodova. Istraživanja reproduktivne biologije predstavnika roda Prunus, korišćenjem različitih metodologija (molekularnih, bioloških, histoloških), obezbedila su značajne informacije o biološkim događajima bitnim za zametanje plodova. Različiti aspekti koji se odnose na vreme cvetanja, potrebu za oprašivačima, muški i ženski gametofit, njihove performanse i interakcije (koje su u osnovi reproduktivnog uspeha) u različitim ekološkim kontekstima, pokazali su se kao veoma bitne teme od značaja za strategije oplemenjivanja i stvaranje novih sorti. Pokazalo se da su određene genetički determinisane reproduktivne osobine, kao što su kratko vreme cvetanja i životni vek ovula kao i, posledično, kratak efektivni polinacioni period, samobesplodnost, za modernu hortikulturnu praksu manje poželjne ili nepoželjne. Rezultati pokazuju da aktuelne klimatske promene imaju negativni uticaj na produkciju plodova u voćarstvu. Razmatrajući  sadašnju situaciju kao osnov za budući razvoj u oblasti oplemenjivanja i proizvodnje plodova koštičavih vrsta voćaka, neophodni su dalji integrativni pristupi. Novi ciljevi oplemenjivanja vrsta roda Prunus moraju objediniti adaptabilnost na temperaturne okolnosti koji proističu iz klimatskih promena, kvalitet ploda i otpornost na bolesti. U tom smislu, proučavanje fiziologije reproduktivnih procesa u narednim godinama će postajati sve značajnije, iz perspektive suočavanja sa promenama uslova životne sredine.
AB  - Considerable progress has been made in the breeding of stone fruits worldwide, in order to obtain cultivars that are adapted to different agro-ecological conditions, and with characteristics that ensure commercial effects and respond to consumer demands for healthy and quality fruits. Studies of the reproductive biology of Prunus species have combined different methodologies (molecular, biological, histological), providing valuable information on biological events essential for optimal fruit set. Different aspects related to blooming time, the needs for pollenisers, male and female gametophytes, their performances and interactions (that underlie reproductive success) under various ecological contexts, were shown to be important topics that contribute to the breeding strategies and developing of the new cultivars. Genetically determined reproductive traits such as short blooming time or ovule longevity and, consequently, insufficient effective pollination period, and self-incompatibility, have been shown to be less desirable or unsuitable from the aspect of modern horticultural practice. Nowadays, results also indicate that ongoing climate change has detrimental effects on fruit production. Considering different aspect of the current situation as a baseline for the future development of the stone fruits breeding and production, the future integrative approaches are necessary. The new breeding objectives of Prunus species must unify adaptability to temperature conditions arising from climate change, fruit quality and disease resistance. In this regard, the study of the physiology of reproductive processes will become increasingly important in future years from the perspective of coping with such environmental changes.

AB  - Pollination and temperature are predominant factors affecting the progamic phase of sexual plant reproduction.
In light of climate change and the emergence of increasing temperatures, new knowledge is necessary to
encourage fruit breeding programmes to develop cultivars adapted to these changes, as well as to find the best
incompatibility relationships with the aim of achieving high fruit quality and yield. This research was conducted
to reveal the effect of pollination treatments and temperature on the progamic phase and fruit set in three European
plum (Prunus domestica L.) cultivars (‘Jojo’, ‘Hanita’ and ‘Katinka’) resistant/tolerant to Sharka virus
using fluorescent microscopy. Reproductive parameters (flowering overlapping, pollen germination, number of
pollen tubes in the upper part of the style and locule of the ovary, dynamics of pollen tubes growth and initial and
final fruit set) were investigated in vivo under different pollination treatments (cross, open and self-pollination).
The influence of different temperature regimes (20 ◦C, 23 ◦C and 26 ◦C) on the progamic phase was also observed
under cross and self-pollination. The pollination treatments significantly influenced the number of pollen tubes in
the style and ovary, as well as dynamics of pollen tubes in different regions of pistil. Pollen tubes showed better
growth in different crossing combinations compared to open and self-pollination. Higher temperatures accelerated
pollen tube growth and reduced the time they need to reach ovules. Generally, ‘Jojo’ appeared to be the
best pollenizer among the examined cultivars, while ‘Katinka’ exhibited the best response of pollen tube growth
to various temperature regimes. The differences observed between growth of the pollen tubes in each genotypetemperature
combination revealed that certain crossing combinations were better adapted to higher temperatures
so this can be taken into account as one of the adjustments to climate change. The ability of cultivars to
respond to high temperatures in the progamic phase can also be helpful for breeding programs in developing new
cultivars better adapted to different temperature conditions.

Identification of S-haplotypes in autochthonous, released and promising sour cherry genotypes originated from the Republic of Serbia

AB  - Sour cherry (Prunus cerasus L.) is one of the main perennial fruit crops in the Republic of Serbia (RS), whose average production of 117,595 tonnes (2012-2021) ranking it as the third most widely grown after plum and apple. As for the sour cherry assortment structure in the RS, clones of ‘Oblačinska’ are the most widespread (over 60%), while the remaining part comprises large-fruited foreign cultivars, as well as domestic ‘Šumadinka’ released within the breeding programme of the Fruit Research Institute, Čačak (FRI). As the tetraploid species, sour cherry includes both self-incompatible (SI) and self-compatible (SC) genotypes, which are characterised by more complex genetics of SI/SC. Therefore, developing of SC sour cherry cultivars is of high priority within FRI breeding work and worldwide. The aim of this study was to identify the S-haplotypes in two autochthonous (clones of ‘Oblačinska’ and ‘Cigančica’), four developed within FRI breeding programme (‘Iskra’, ‘Nevena’, ‘Sofija’ and ‘Šumadinka’) and one promising (‘GV-10’) sour cherry genotypes originated from the RS. The use of the polymerase chain reaction (PCR) method with three different consensus primer pairs for the first and the second introns of S-RNase, as well as primers specific for both S-RNase and SFB alleles, in combination with genetic tests to identify the pollen-part and stylar-part mutants (using derived cleaved amplified polymorphic sequence assay ‒ dCAPS), enabled identification of the S-haplotypes in the assessed sour cherry genotypes. In total, eight S-haplotypes (S1, S4, S6, S9, S13, S26, S35 and S36) or their non-functional variants were identified. In the investigated sour cherry genotypes, non-functional variant S36b2 was the most frequent (21.4%). The results generated in this study provide a valuable resource for design crosses in future breeding work which maximize the number of SC seedlings, and for orchard management in the efficient high-yielding fruit production.

AB  - The study was carried out to determine the effect of the site and temperature regime on the in vitro pollen performance of sweet cherries ʻLapins’, and ʻBurlat’, and sour cherry ʻŠumadinka’ in order to evaluate their ability as pollen donors for some Serbian cherry cultivars. For each cultivar pollen was collected from the two sites (Beograd and Čačak). In vitro pollen performance was conducted at 17°C and 20°C. Pollen germination was determined in a medium containing 0.7% agar-agar and 15% sucrose. For sweet cherries, increasing the temperature from 17°C to 20°C resulted in a significant decrease in pollen germination and pollen tube growth, while for ʻŠumadinka’ opposite result, but without a significant difference was observed. A significant influence of the site on pollen germination was established. Higher values were recorded for both sweet cherries in Čačak, while ʻŠumadinka’ showed a better response in Beograd. The shortest pollen tubes were observed in Burlat (370.6μm) while ʻŠumadinka’ had the longest ones (830.5μm). Site and temperatures affected pollen tube growth in vitro. Different germination percentages and pollen tube growth were found among both, different cultivars located in the same place and the same cultivars from two different locations. This leads to the different environmental conditions that influenced pollen formation in each location. More evaluation, including pollen performance in vivo and fruit settings in cross-pollinated combination with Serbian cherry cultivars is needed for the proper choice of pollen donors.

AB  - Lack of appropriate fertilisers is a bottleneck in organic agriculture, which can lead to reduced yields and, consequently, reduced profitability. Therefore, the aim of this study was to develop a new formulation of a vermicompost-based biopreparation (VCMo) and to investigate the effects of this product on the composition of primary and secondary metabolites of the fruit of organically cultivated strawberries (Fragaria × ananassa Duch. cv Senga Sengana). Sugars, organic acids and phenolic compounds in strawberry fruit cultivated with three application modes of VCMo (rhizosphere inoculation (RI); combination of rhizosphere inoculation and foliar spraying (RI+FS); and foliar spraying (FS)) and a control (untreated soil and plants) were characterised and quantified by HPLC with a refractive index detector for sugars and organic acids, and HPLCDAD-MSn for phenolic compounds. Strawberry fruit obtained from plants exposed to the combined application of rhizosphere inoculation and foliar spraying showed the highest concentrations of total sugars and total organic acids. There were no significant differences between the methods of VMCo application for concentrations of phenolic acids, but rhizosphere inoculation on its own resulted in the highest total concentration of the five groups of flavonoids identified (anthocyanins, flavanols, flavanones, flavonols and flavones). The results of the study suggested that rhizosphere inoculation with VCMo on its own or in combination with foliar spraying could be а useful tool for organic strawberry cultivation, contributing to the production of healthy and good quality fruit at the same time as providing opportunities to reduce environmental pollution.

U zasadima šljive u Srbiji je zastupljen određen broj autohtonih sorti šljive čiji se plodovi prvenstveno koriste za proizvodnju rakije. Jedna od njih je i ‘Crvena ranka’ koja predstavlja najznačajniju i najviše zastupljenu autohtonu sortu šljive. Karakteriše je samobesplodnost, odnosno pojava citoplazmatične muške sterilnosti, pa je stoga u zasadima ove šljive neophodno prisustvo adekvatnog oprašivača. Cilj ovog rada je utvrđivanje efektivnog polinacionog perioda (EPP) kod ove sorte, kroz ispitivanje broja inicijano i finalno zametnutih plodova. Tokom proleća 2021. godine, od momenta punog cvetanja pa do desetog dana nakon toga, svaki dan je izvršeno oprašivanje oko 200 cvetova, emaskuliranih u fazi poznog balona. Za oprašivanje je korišćena smeša suvog polena sorti šljive ‘Čačanska lepotica’, ‘Čačanska rodna’ kao i autohtone sorte ‘Dragačica’. Nakon dve do tri nedelje od oprašivanja utvrđen je broj inicijalno zametnutih plodova, a finalno u periodu fiziološke zrelosti ploda. Najveći broj zametnutih plodova dobijen je oprašivanjem cvetova u prvih šest dana od početka punog cvetanja, odnosno od trećeg do šestog dana. Nakon šestog dana dolazi do pada broja zametnutih plodova, da bi kod cvetova oprašenih desetog dana od momenta punog cvetanja vrednosti broja inicijalno i finalno zametnutih plodova bile 17% i 11%. Na osnovu dobijenih rezultata EPP kod ove sorte je trajao sedam dana.
AB  - The area of the Balkan Peninsula is characterized by the presence of a large number of autochthonous plum cultivars with different local names and most often of unknown origin. A number of autochthonous plum cultivars whose fruits are primarily used for the brandy production are represented in plum orchards in Serbia. One of them is ‘Crvena Ranka’, which is the most important and most represented autochthonous plum variety. It’s a self-sterile cultivar (characterized by the appearance of cytoplasmic male sterility), so the presence of an adequate pollenizer is needed. This study aimed to examine the effective pollination period of this cultivar, based on the initial and final fruit set. During spring 2021, from the moment of full flowering until the tenth day after that, about 200 flowers (emasculated in the late "balloon" phase) were pollinated every day. A mixture of dried pollen from the plum cultivars ‘Čačanska Lepotica’, ‘Čačanska Rodna’ as well as the autochtonous cultivar ‘Dragačica’ was used for pollination. Three to four weeks after pollination, the initial fruit set was determined while the final fruit set was determined at the period of physiological maturity of the fruit. The highest percentage of fruit set was obtained by pollination of flowers during the first six days from the beginning of full flowering, ie from the third to the sixth day. In the flowers pollinated after the sixth day from the full flowering, a decrease in the fruit sets was observed, with the lowest value on the tenth day from the date of full flowering amounted of 17% for initial and 11% final fruit set. AB  - In addition to conventional breeding which has been resulted in release of two sweet cherry (P. avium) and five sour cherry (P. cerasus) cultivars, different aspects of reproductive biology in cherries are among the main research activities at Fruit Research Institute, Čačak (FRI). The most important results were obtained in the field of pollination and fertilization of cherries – flowering phenology, pollen quality, monitoring the pollen tube growth in vitro and in vivo (pollen tube number and growth rate through certain pistil parts, pollen-pistil interaction in the style, unusual pollen tube growth in the ovary), cytoembryology (stage of ovule development, viability of ovule and embryo sac, early embryogenesis). Investigations were also focused to specifities of cherry genotypes in their reproductive behaviour as polleniser/pollinated cultivar, and to the adequate choice of domestic and foreign cultivars' combinations that exhibit the best performance in terms of fruit set and yields. In the last decade, research work on self- and cross-(in)compatibility in sweet cherry has been considerably advanced by S-genotype identification of cultivars, landraces and hybrids using the consensus and specific primers to amplify the S-RNase alleles. Nowdays, pollen-pistil interactions have been considered in the context of temperature conditions, bearing in mind incidence of seasons with higher temperatures during the flowering in the main cherry-growing regions. Within the national CherrySeRB project whose FRI is the leading institution, characterisation of the indigenous genotype potential for desirable reproductive properties and defining ʻgood reproductive behaviour cherry modelʼ that can face warmer temperature conditions during the flowering are in focus. The main idea of CherrySeRB is connection of production challenges arising from climate change and the richness of Serbian and Balkan cherry germplasm that is still unused enough, which represents a novelty in breeding approach, applicable to other fruit species

AB  - Grey mould (Botrytis cinerea Pers.) is one of the most important strawberry diseases, which in conventional farming is successfully controlled by chemical fungicides during flowering. Yet, in organic farming, due to the unavailability of appropriate non-fungicidal methods, its control is one of the biggest problems that can often cause a significant reduction in production and profitability. Thus, the study is aimed to investigate the possibility of using some biological alternatives (soil microorganisms) for controlling grey mould disease in strawberry plantings. For that purpose, in Fruit Research Institute Čačak (Republic of Serbia) a new formula of biopreparation (VCMo) consisting of microbes from Trichoderma, Bacillus, Pseudomonas and Azotobacter genera has been developed. The VCMo was applied at the beginning, full and the end of flowering stage. In three harvest times (beginning, middle and the end), its efficiency has been investigated by controlling B. cinerea of the fruits of organically cultivated ʻAlbaʼ strawberries (Fragaria x ananassa Duch). Besides, yield, physical fruit properties and soluble solids have been monitored also. Strawberry fruits obtained from plants exposed to VCMo application showed a lower incidence of grey mould (38.29%). In addition, degree of infection has been decreased from the beginning till the end of harvest (55.08, 49.97 and 39.13%, respectively). Higher yield of usable fruits was observed under VCMo application, especially in the middle harvest time (148.31 g/plant). The fruit weight and dimensions, as expected, varied under the influence of the harvest date. The application of VCMo showed a positive effect on the fruit weight in the middle and at the end of the harvest (28.61, 20.81 g), which can also be said for soluble solids (5.88 and 6.58 oBrix). Contrary, the fruit firmness was the highest at the beginning of the harvest (1.47 kg cm2).

AB  - A study was conducted to investigate the effect of different storage periods and temperatures on pollen viability in vitro and in vivo in plum genotypes 'Valerija', 'Cacanska Lepotica' and 'Valjevka'. In vitro pollen viability was tested at day 0 (fresh dry pollen) and after 3, 6, 9 and 12 months of storage at four different temperatures (4, -20, -80 and -196 degrees C), and in vivo after 12 months of storage at distinct temperatures. In vitro germination and fluorescein diacetate (FDA) staining methods were used to test pollen viability, while aniline blue staining was used for observing in vivo pollen tube growth. Fresh pollen germination and viability ranged from 42.35 to 63.79% ('Valjevka' and 'Cacanska Lepotica', respectively) and 54.58 to 62.15%, ('Valjevka' and 'Valerija', respectively). With storage at 4 degrees C, pollen viability and germination decreased over the period, with the lowest value after 12 months of storage. Pollen germination and viability for the other storage temperatures (-20, -80 and -196 degrees C) were higher than 30% by the end of the 12 months. Pollination using pollen stored at 4 degrees C showed that pollen tube growth mostly ended in the lower part of the style. With the other storage temperatures, pollen tube growth was similar, ranging between 50 and 100% of the pistils with pollen tubes penetrated into the nucellus of the ovule in the genotype 'Cacanska Lepotica'. The results of these findings will have implications for plum pollen breeding and conservation.

With storage at 4 degrees C, pollen viability and germination decreased over the period, with the lowest value after 12 months of storage. Pollen germination and viability for the other storage temperatures (-20, -80 and -196 degrees C) were higher than 30% by the end of the 12 months. Pollination using pollen stored at 4 degrees C showed that pollen tube growth mostly ended in the lower part of the style. With the other storage temperatures, pollen tube growth was similar, ranging between 50 and 100% of the pistils with pollen tubes penetrated into the nucellus of the ovule in the genotype 'Cacanska Lepotica'. The results of these findings will have implications for plum pollen breeding and conservation.",

AB  - Timočanka je visoko kvalitetna stona sorta šljive, koja se pored pozitivnih osobina kao što su krupan i veoma ukusan plod, odlikuje i promenljivom rodnošću. U cilju analize reproduktivnog procesa ove sorte tokom 2016. i 2017. godine proučavana je klijavost polena in vitro, kao i rast polenovih cevčica in vivo, inicijalno i finalno zametanje plodova u varijantama slobodnog-, samo- i stranooprašivanja (za koje su korišćene sorte Čačanska lepotica, Čačanska rodna i Stanley). Pored toga, eksperimentom je bilo obuhvaćeno i proučavanje fenofaze cvetanja i dnevnih temperatura vazduha, proučavanje vitalnosti embrionove kesice kod neoprašenih cvetova i uspeha oplođenja kod cvetova uzetih iz varijante slobodnog oprašivanja u prvih 21 dan nakon punog cvetanja. Utvrđeno je da su korišćeni oprašivači zadovoljavali kriterijume u pogledu preklapanja feno faze cvetanja sa sortom timočanka, kao i vitalnosti polena. Srednja temperatura za periodod 21 dan nakon punog cvetanja u 2016. Godini iznosilaje 15,3°C, odnosno 11,3°C u 2017. godini. Najviše vrednosti kvantitativnih parametara rasta polenovih cevčica in vivo utvrđene su u varijanti oprašivanja polenom sorte Čačanska lepotica, dok su najviše vrednosti inicijalnog (32,15%) i finalnog zametanja plodova (20,52%) utvrđene u varijanti oprašivanja polenom sorte Čačanska rodna. Najniže vrednosti proučavanih parametara utvrđene su u varijanti samooprašivanja, kao i u varijanti stranooprašivanja polenom sorte Stanley. Svi proučavani parametri polena, kao i inicijalno i finalno zametanje plodova, imali suviše vrednosti u prvoj godini proučavanja. Vitalne embrionove kesice su u obe godine zapažene do desetog dana posle punog cvetanja (16,7% u 2016. i 27,3% u 2017). Međutim, broj oplođenih embrionih kesica bio je veći u 2016. godini u odnosu na 2017. Utvrđena je i značajna pozitivna korelacija između vitalnosti polena, kvantitativnih parametara rasta polenovih cevčica in vivo i zametanjap lodova.
AB  - The dessert plum cultivar ‘Timočanka’, in addition to positive characteristics such as large fruits of excellent taste and good quality, is also characterized by irregular bearing. In order to evaluate the reproductive process during two years (2016 and 2017), in vitro pollen germination, quantitative parameters of pollen tube growth in vivo, initial and final fruit set under open, self- and cross-pollination (with cultivars ʽČačanska Lepoticaʼ, ʽČačanska Rodnaʼ and ʽStanleyʼ) were evaluated. Also, the experiment included examination of the flowering phenophaseand daily air temperatures, embryo sac viability in unpollinated flowers and fertilization success in flowers from open pollination within the 21 days following the date of full flowering. The results indicated that pollenizers had good pollen germination in vitro and satisfactory flowering time overlap with the cultivar ʽTimočankaʼ. Mean daily temperatures for flowering and post-flowering period in 2016 and 2017 were 15.3°C and 11.3°C, respectively. The highest average values of quantitative parameters of pollen tube growth in vivo were obtained in crosspollination variant with ʽČačanska Lepoticaʼ, while the highest average values of initial (32.15%) and final (20.52%) fruit set were observed with ʽČačanska Rodnaʼ as pollenizer. The lowest values of examined parameters were recorded in the self-pollination variant and in the cross-pollination variant with cultivar ʽStanleyʼ as pollenizer. All investigated parameters of pollen tube growth, as well as initial and final fruit set had higher values in the first experimental year. In both years during the first ten days after full bloom, the viable embryo sacs were observed (16.7% in 2016 and 27.3% in 2017). However, fertilization success i.e., number of fertilized embryo sacs was higher in 2016. than in 2017. The study revealed existence of significant correlations between individual studied parameters of pollen viability, pollen tube growth in vivo and fruit set.

AB  - Poljoprivrednu proizvodnju poslednjih decenija prati degradacija životne sredine koja ugrožava same sisteme od kojih zavisi naš budući razvoj. Pored toga, konvencionalni sistemi poljoprivredne proizvodnje dostigli su svoj maksimum u smislu povećanja produktivnosti i korišćenja zemljišta. Shodno tome, istraživanja u oblasti biotehnologije postaju sve aktuelnija, a pronalaženje odgovarajućih eko-bezbednih preparata i uvođenje inovativnih i po životnu sredinu bezbednih tehnika u voćarsku proizvodnu praksu predstavljaju veliki izazov za naučnike širom sveta. Optimizacija procesa dobijanja mikrobne biomase, kao i tehnološkog postupka formulacije mikrobioloških preparata sa potencijalnim biopesticidnim i fitostimulatornim efektom predstavljaju odličnu alternativu i veliku šansu koja može odgovoriti gore pomenutim izazovima. 
U radu su opisana najnovija iskustva i rezultati bazirani na korišćenju prirodnih resursa i razvoju novih biotehnoloških rešenja koja mogu podstaći rast i prinos voćaka i poboljšati nutritivnu vrednost plodova. U tom smislu, posebna pažnja je posvećena rezultatima dosadašnjih proučavanja efikasnosti primene jednog inovativnog biopreparata na bazi vermikomposta, obogaćenog odabranim sojevima korisnih mikroorganizama (BioVermix), razvijenog u Institutu za voćarstvo Čačak. Akcenat je stavljen na mehanizame njegovog dejstva i praktične primere njegove primene, kao nove, bioracionalne metode u voćarskoj proizvodnoj praksi.
Rezultati sprovedenih istraživanja pokazali su da primena BioVermix-a u zasadima šljive, jagode, aronije, kupine i borovnice omogućava postizanje stabilnih prinosa i visok kvalitet ploda sa povećanim sadržajem fenolnih jedinjenja odgovornih za antioksidativni kapacitet. Konačno, sve ovo zajedno omogućava kreiranje sistema input-primarni-finalni proizvod bez negativnih posledica na ljudsko zdravlje i životnu sredinu.
AB  - Agricultural production in recent decades has been accompanied by environmental degradation that affects the very systems on which our future development depends. Besides, conventional agricultural production has reached its maximum in terms of increasing productivity and land use. Accordingly, research in the field of biotechnology is becoming more and more relevant, and discovering appropriate eco-safe preparations and introducing innovative and environmentally safe techniques into fruit growing is a major challenge for scientists around the world.Optimization of the production of microbial biomass and formulation of microbiаl preparations with potential biopesticidal and phytostimulatory action are an excellent alternative which may respond to the challenges mentioned above. In this regard, primarily due to the emergence of environmental awareness of consumers and as well as human health.
The study describes the latest experiences and results based on the use of natural resources and development of new biotechnological solutions that can stimulate fruit growth and yielding and improve the nutritional value of fruits as well. In that sense, special attention is paid to the results of previous studies on the efficiency of one innovative biopreparation based on vermicompost, enriched with selected strains of beneficial microorganisms (BioVermix), developed at the Fruit Research Institute, Čačak. Emphasis is placed on the mechanisms of its action and practical examples of its application, as new, biorational methods in fruit growing.
The results of the research showed that the application of BioVermix in plum orchards and plantings of strawberries, chokeberries, blackberries and blueberries enables stable yields and high quality fruits with increased content of phenolic compounds responsible for antioxidant capacity. Finally all this together enables fruit growers to create of an input-primary-final product system without negative effects on human health and the environment.

Volatile components in fruits of raspberry cultivars and selection grown in Western Serbia

AB  - This study presents the results of a research into volatile compounds in raspberry fruits of four commercial cultivars (‛Willamette’, ‛Meeker’, ‛Tulameen’, ‛Latham’) and one a selection (‛K 81/6’) grown in Western Serbia. By using the method of simultaneous distillation and extraction (Likens-Nickerson method), 37 volatile compounds were isolated and identified (using gas chromatography-mass spectrometry – GC/MS), and then classified into corresponding classes of aldehydes, ketones, acids, esters, terpenes, C13-norisoprenoids, sesquiterpenes and hydrocarbons. A quantitative analysis (using gas chromatography-flаmе iоnisаtiоn dеtеctоr – GC/FID) revealed that tested raspberry cultivars and selection differ in the content of individual volatile components (expressed in %). The highest share in volatile components belonged to the class of C13-norisoprenoids, among which the most abundant were β-ionone (ranging from 4.50% in ‛Latham’ to 26.79% in ‛K 81/6’ selection) and α-ionone (ranging from 5.04% in ‛Latham’ cv., to 11.43% in ‛Meeker’).
AB  - Prоizvоdnjа mаlinе u Srbiјi u prеdhоdnih pеt gоdinа krеtаlа sе izmеđu 42.300 i 100.000 tоnа (prоsеk 70.400 tоnа), što svrstаvа našu zemlju u rеd vоdеćih svetskih prоizvоđаčа mаlinе. Zаpаdnа Srbiја је glаvnо prоizvоdnо pоdručје zа gајеnjе mаlinе. Nа izbоr sоrtе mаlinе kоја ćе sе gајiti nа оdrеđеnоm pоdručјu, pоrеd njеnih оdgајivаčkih kаrаktеristikа i prilаgоđеnоsti datim аgrо-еkоlоškim uslоvimа, prеsudnо utiču kvаlitеt i nаčin kоrišćеnjа plоdа. Kvalitet plodova se najčešće povezuje sa sеnzоrnim kаrаktеristikаmа plоdа (izglеd, kоnzistеnciја, ukus i аrоmа) koje zavise od brојnih kоmpоnеnata hеmiјskоg sаstаvа. Od ispаrlјivih kоmpоnеnаtа plоdа mаlinе zаvisi njеnа аrоmа, odnosno оvа јеdinjеnjа, zајеdnо sа šеćеrimа i kisеlinаmа, dајu spеcifičаn miris i ukus mаlinе. Оvе sеnzоrnе kаrаktеristikе su pаrаmеtri kvаlitеtа оd kојih, izmеđu оstаlоg, zаvisi prihvаtlјivоst plоdоvа rаzličitih sоrаtа mаlinе zа pоtrоšnju u svеžеm stаnju, kао i pоgоdnоst zа prеrаdu. Pošto se gotovo celokupan godišnji rod malina u Srbiji prerađuje, zа dоbiјаnjе visоkоg kvаlitеtа pојеdinih prоizvоdа, vаžnо је kоristiti sоrtе mаlinе kоје sе kаrаktеrišu, izmеđu оstаlоg, i izrаžеnоm karakterističnom аrоmоm.  
U rаdu su prikаzаni rеzultаti ispitivаnjа isparljivih mаtеriја plоdоvа čеtiri sоrtе (Vilаmеt, Мikеr, Тјulаmin, Lејtаm) i јеdnе sеlеkciје (K 81/6) mаlinе gajenih u Zapadnoj Srbiji. Меtоdоm simultane destilacije i ekstrakcije (metod pо Lickens-Nickerson-u) izоlоvаnо је i identifikovano 37 аrоmаtičnih kоmpоnеnаtа, kоје su nаkоn idеntifikаciје (mеtоd GC/MS) svrstаnе u klase aldehida, ketona, kiselina, estara, terpena, C13 norizoprenoida, sеskvitеrpеnа i uglјovodonika. Među identifikovanim isparljivim jedinjenjima, 16 je nađeno u plodovima svih pet genotipova. Оstаlе kоmpоnеntе bilе su prisutnе  u plоdоvimа sаmо pојеdinih gеnоtipоvа. Меđu ispаrlјivim јеdinjеnjimа kаrаktеrističnim zа svе gеnоtipоvе, 11 kоmpоnеnаtа spаdајu u grupu visоkо аrоmаtski pоtеntnih јеdinjеnjа i tо su 4 tеrpеnа (α-pinеn, limоnеn, linаlоl, α-tеrpinеоl), 4 C13-nоrizоprеnоidа (α-јоnоl, dihidrо α-јоnоn, α-јоnоn, β-јоnоn) i 3 аldеhidа (nоnаnаl, dеkаnаl, ciklаmа аldеhid). 
Kvаntitаtivnоm аnаlizоm (GC/FID) је utvrđеnо dа sе plоdоvi ispitivаnih sоrаtа i sеlеkciје mаlinе rаzlikuјu pо sаdržајu pојеdinih kоmpоnеnаtа аrоmе (izrаžеnih u %). Nајzаstuplјеniје kоmpоnеntе аrоmе plоdоvа pripаdаlе su klаsi C13 nоrizоprеnоidа, оd kојih su nајzаstuplјеniјi bili β-јоnоn (оd 4,50% kоd sоrtе Lејtаm dо 26,79% kоd sеlеkciје K 81/6) i α-јоnоn (оd 5,04% kоd sоrtе Lејtаm dо 11,43% kоd sоrtе Мikеr).
Nа оsnоvu hiјеrаrhiјskе klаstеr аnаlizе ispitivаni gеnоtipоvi mаlinе sе mоgu svrstаti u dvа klаstеrа. U јеdnоm klаstеru sе nаlаzе gеnоtipоvi čiјi plоdоvi imајu izrаžеnu kаrаktеrističnu аrоmu mаlinе (Vilаmеt, Мikеr, Тјulаmin i K81/6), uz pоstојаnjе finih gеnоtipskih rаzlikа. U drugоm klаstеru sе nаlаzi sоrtа Lејtаm, čiјi је prоfil аrоmе pоtpunо drugаčiјi оd оstаlih gеnоtipоvа.

abstract = "This study presents the results of a research into volatile compounds in raspberry fruits of four commercial cultivars (‛Willamette’, ‛Meeker’, ‛Tulameen’, ‛Latham’) and one a selection (‛K 81/6’) grown in Western Serbia. By using the method of simultaneous distillation and extraction (Likens-Nickerson method), 37 volatile compounds were isolated and identified (using gas chromatography-mass spectrometry – GC/MS), and then classified into corresponding classes of aldehydes, ketones, acids, esters, terpenes, C13-norisoprenoids, sesquiterpenes and hydrocarbons. A quantitative analysis (using gas chromatography-flаmе iоnisаtiоn dеtеctоr – GC/FID) revealed that tested raspberry cultivars and selection differ in the content of individual volatile components (expressed in %). The aim of the present work was to study the development and viability of embryo sacs, as well as fertilization success, in relation to the fruit set of the cultivars 'Mallard', 'Edda', 'Jubileum', and 'Reeves', under specific Norwegian climatic conditions. Emasculated, unpollinated, and open-pollinated flowers were collected at the beginning of flowering, and on the 3rd, 6th, 9th, and 12th days after flowering, from all four plum cultivars over two years (2018/2019). Ovaries were dehydrated, embedded in paraffin wax, sectioned, stained, and observed under a light microscope. Results showed the existence of synchronization between successive phases in the development of the embryo sac and individual phases of flowering. All plum cultivars had higher percentages of viable embryo sacs, fertilized embryo sacs, and fruit set in 2018 than in 2019. These differences may be related to the very low temperatures during the post-full-flowering period in 2019, and to the low adaptation of some studied cultivars to unfavorable conditions. In our study, the cultivar 'Jubileum' showed the highest percentage of viable embryo sacs, fertilized embryo sacs, and fruit set compared to other cultivars, i.e., the best low-temperature adaptation.

Assessment of self-(in)compatibility in new plum (Prunus domestica L.) cultivars developed at Fruit Research Institute, Čačak

AB  - Breeding work at Fruit Research Institute, Čačak is predominantly focused on developing new European plum (Prunus domestica L.) cultivars. Since its establishment until now, 18 plum cultivars have been named and released. Knowledge of self-(in)compatibility status of these cultivars is of great importance for growers to design orchards, and for breeders to choose parents in future breeding programmes. The objective of this study was to determine the degree of self-(in)compatibility of three new plum cultivars released in 2018 (‘Petra’ and ‘Divna’) and 2020 (‘Lana’). The investigation was conducted during two years (2018/19), and included pollen performance in vitro (germination on agarose-sucrose medium) and in vivo (the number of pollen tubes in the upper third of the style, at the base of the style and in the ovary; the percentage of pistils with the pollen tube penetrating the nucellus 10 days after pollination), as well as examination of fruit set under self- and open pollination modes. In vitro pollen germination had high values, ranging between 61.57% (‘Lana’) and 62.73% (‘Petra’). ‘Divna’ and ‘Petra’ were characterized by the high values of all parameters of pollen tube growth in vivo, as well as fruit set in both self-pollination (33.26 and 30.35%, resp.) and open pollination (53.18% and 48.74%, resp.) variants. In the pistils of ‘Lana’, pollen tubes growth inhibition was manifested in the upper parts of the style after self-pollination, so in the base of the style and in the ovary no tubes were noticed; fruit set was not observed as well. Somewhat lower values of all mentioned parameters (except the number of pollen tubes in the upper third of the style) were also found in ʻLanaʼ under open pollination mode. Based on the obtained results, ‘Lana’ can be characterized as self-incompatible, while ‘Petra’ and ‘Divna’ are self-compatible cultivars.

Success rate of individual pollinizers to European plums

AB  - Fruit set and yield optimization in orchards depend on the success of pollination and fertilization. There is a wide variation in the self-fertility among European plum genotypes from self-fertile to completely self-incompatible requiring compatible pollinizers with overlapping flowering times. Thus, for the successful cultivation it is necessary to determine the best pollinizers in order to obtain high yields. The aim of this investigation was to find the most suitable pollinizers for the cultivars ‘Edda’, ‘Mallard’, ‘Reeves’ and ‘Jubileum’, which are well adapted to the production in the Nordic climate, and in order to recommend to the producers which cultivars should be planted within the same orchard to provide large and stable, annual yields. The phenology of flowering of these plum cultivars was monitored during the springs 2018 and 2019 at different plum growing regions. Pollen viability, cross pollination with different pollinizers included open- and self-pollination, pollen tube growth and pollen–pistil incompatibility reactions were investigated. The dynamics of pollen tube growth (third, sixth and ninth day after anthesis) in the style (upper, middle and lower third) and parts of ovary in different crossing combination were observed by fluorescent microscopy over the same two years in relation to fruit set. Overview and preliminary results from the different experiments will be presented.

AB  - The aim of this study was to evaluate the content and profile of the phenolic compounds (PCs) and antioxidant properties of field-grown leaves, in vitro leaves and in vitro callus cultures of the blackberry 'Cacanska Bestrna' and blueberry 'Toro'. In vitro shoots of the selected genotypes were grown either on original Murashige and Skoog (MS) medium containing 1 mg/L BA, 0.1 mg/L IBA and 0.1 mg/L GA(3) ('Cacanska Bestrna') or on MS medium with macroelements reduced to 1/2, 2 mg/L zeatin and 0.2 mg/L IAA ('Toro'). Callus cultures were induced from in vitro leaves and established on MS medium with 2 mg/L BA and 2 mg/L 2,4-D ('Cacanska Bestrna') or MS medium with half strength macroelements, 2 mg/L BA, 2 mg/L 2,4-D and 1 mg/L NAA ('Toro'). Total phenolic (TPC) and flavonoid content (TFC) were the highest in blueberry leaves, whereas low TPC and TFC values were obtained in callus cultures of both cultivars. A higher content of PCs in blueberry leaves compared to blackberry leaves was determined by the UHPLC-DAD MS/MS technique. Quercetin derivatives and phenolic acids were the dominant PCs in the leaves of both berries, whereas gallocatechin was present in a significant amount in blueberry leaves. Callus cultures of both berries had a specific PC profile, with none detected in the leaves except quercetin-3-O-glucoside and quercetin-3-O-rutinoside. Blackberry leaves showed the best antioxidant properties as estimated by ferric reducing power (FRP), ABTS(& BULL;+) and DPPH & BULL; scavenging activity assays. Callus cultures of both berries exhibited three to five times lower ABTS(& BULL;+) and ten to seventeen times lower DPPH & BULL; scavenging activity compared to corresponding leaves. The analyzed leaves and callus cultures can be a good source of PCs with good antioxidant properties and specific phenolics, respectively, for applications in the food and pharmaceutical industries.

Reproductive ability of plum (Prunus domestica L.) pollen stored at low temperatures

AB  - Breeding programmes often require the necessity for longer pollen storage, whether asynchronous flowering of parental pairs exists (especially if early-flowering genotypes are pollinated with late-flowering), or if there is a temporal, geographical or spatial distance. The objective of this study was to assessed pollen vitality in vitro and in vivo in three plum cultivars (‘Čačanka Lepotica’, ‘Valjevka’ and ‘Valerija’) after one year of storage at +4°C and sub-freezing temperatures (-20°C, -80°C and -196°C). Pollen viability in vitro was determined on germination medium and by staining with fluorescein-diacetate. For in vivo vitality assay, stored pollen was used for pollination of emasculated flowers of ‘Čačanka Lepotica’. Parameters of pollen tubes growth was determined on 3th, 6th and 10th day after pollination. In all cultivars, the lowest rate of pollen viability was observed for that stored at +4°C. Pollen stored at sub-freezing temperatures showed relatively high viability that was reduced from 8.85 to 47.15% in comparison with fresh pollen. Generaly, in vitro germination test showed lower pollen viability compared to fluorescein-diacetate staining method. Pollen viability under in vivo conditions, corellated with the those obtained under in vitro conditions. The growth of pollen tubes, from pollen stored at +4°C, or was absent or it ended mainly in the upper parts of the style. For pollen stored at sub-freezing temperatures, pollen tubes penetrated into the nucelus on the 6th day after pollination in all cultivars. The best dynamic of pollen tube growth of ‘Čačanska Lepotica’ and ‘Valerija’ was observed with pollen stored at -80°C, while in ‘Valјevka’ with that stored at -196°C.

AB  - ‘Čačanska Rana’ is a dessert plum cultivar of excellent quality that can reach good price in the market. It is self-incompatible cultivar which has irregular yielding with year-to-year variation. For achieving good fruit set and high yields, it is necessary to conduct successful pollination and fertilization where many cyto-embryological processes must take place at the right time. In this study, functionality of embryo sac − their viability and fruit set, were used to evaluate fruit set in open pollination and emasculated unpollinated flowers as control (onset of full bloom on the third, sixth, tenth, fourteenth and twenty-first day after onset of full bloom) in ‘Čačanska Rana’ during the two years period (2015/2017). Considering the results of functionality of embryo sacs (8-, 5- and 4- nucleate stages), their viability was slightly longer in 2015 compared to 2017. In both years, fast degeneration of one out of two synergids and presence of 3- nucleate stage (with normal egg cell) up to fourteenth days of full flowering were observed. The embryo development recorded after fourteenth day after onset of full bloom. On the other hand, fruit set (24%) was higher in 2015, compared to fruit set (5.3%) in 2017. These results indicated that functionality of embryo sac as one of the factors that may be responsible for irregular yield of ‘Čačanska Rana’.

Occurrence of specific behaviour of growing pollen tubes in the ovary of the 'Pozna Plava' plum cultivar

AB  - Plum cultivar 'Pozna Plava' was developed at the Fruit Research Institute, Cacak, and released as a new cultivar in 2008. This plum cultivar bears poorly and irregularly, resulting in very low fruit set. Therefore, the occurrence of specific behaviour of growing pollen tubes in the ovary was studied over a three-year period, in three pollination variants (self-, cross- and open). The cross-pollination was performed using the pollen of 'Cacanska Najbolja', 'Hanita' and 'Presenta' plum cultivars. Growth of pollen tubes was observed by means of fluorescent microscopy. The analysis was conducted taking into consideration whether or not the pollen tube has penetrated into the nucellus. Presence of specific behaviour of growing pollen tubes in the ovary was observed in all pollination variants. Atypical pollen tube growth was recorded in the locyle of the ovary as well as in the region of the obturator, the micropyle and the embryo sac. The largest percentage of atypical pollen tube growth in all pollination variants, before and after the penetration into the nucellus was recorded in the obturator and micropyle region. In the open pollination variant, the ovaries showing this specific behaviour of growing pollen tubes, were noted in only one year. Cross- and self-pollination variant showed the existence of specific behaviour of growing pollen tubes in all regions of the ovule, including the embryo sac (except in self-pollination). Only in cross-pollination variant, correlation-regression analysis has shown that specific behaviour of growing pollen tubes did affect the pollen tube penetrating into the nucellus.

Fertilization biology of 'Reka' highbush blueberry

AB  - The paper presents results of the three-year study of fertilization biology of 'Reka' highbush blueberry (Vaccinium corymbosum L.) under the cross-, open and self-pollination variants. Cross-pollination was performed using the pollen of 'Bluecrop', 'Duke', 'Nui' and 'Ozarkblue'. Pollen tube growth in vivo was monitored using fluorescent microscopy method. The folowing parameters were observed: the average number of pollen tubes in the upper part and base of the style, number of ovary locules with the penetration of pollen tubes, and polen tubes' growth dynamics through certain pistil parts. The highest number of pollen tubes in the upper part and base of the style, as well as the fastest penetration into the ovary locules were found in the open pollination variant. Significant decrease of pollen tubes number from the upper third to the base of the style has been recorded in all pollination variants, whereas in the self-pollination variant it was to the greatest extent. The results also indicated that pollen tube growth dynamics in the progamic phase of fertilization was significantly influenced by pollenizer genotype and environmental factors, temperature in particular. On the second day after pollination, pollen tubes penetrated the ovary locules in all pollination variants, implying that favourable period for fertilization in this cultivar was one to two days after flower opening. In self- and cross-pollination variants, dynamics of pollen tubes growth was similar, indicating that cultivar 'Reka', commonly grown in multi-varietal, could also be grown in mono-varietal plantations (without pollenizers).
